Objective:The purpose of this study was to investigate the mechanism of the Qinxiang granules on Human Rotavirus diarrhea by observation on the effects of Qinxiang granules on IFN-γand IL-2 of suckling mice infected with Human Rotavirus (HRV),in order to provide more scientifically theoretical and experimental evidence for its clinical application.Methods:Sucling mice were infected by HRV through the mouth and randomly divided into 5 treated groups and one model group,which differently treated with Qinxiang granules (High dose/medium dose/low dose), Ribavirin,Changkang tablets and NS,3 times a day, for 5 days. A separated control group was set and treated as the model group. Suckling mice in each group observed the general situation; decapitation and collected blood, using ELISA assay to observe the changes of serum IFN-γ, IL-2 level; using colloidal gold methods to detecte HRV antigen stool; HE staining under light microscope observations of small intestinal mucosal cells in pathological changes.Results:①12h after modeling, colloidal gold method detected HRV antigen in the model group as positive and normal control group was negative;②general observation:the model group's activity was reducing,diarrhea, weight grow slowly or decline; normal group without diarrhea, weight increased rapidly. Compared with model group, the suckling mice in treatment groups were generally better, of which Qinxiang high-dose group has the lightest level of diarrhea.③Rat feces HRV antigen seroconversion rates in each group:model group,0%; Ribavirin group,87.5%;Qinxiang high-dose group,75%;Qinxiang middle-dose group,62.25%;Qinxiang low-dose group,37.5%;Changkang tablet group,62.5%,there was no significant difference between the sixs groups (P>0.05).④Serum INF-y level:Compared with normal group, the other six infected groups shown significantly increased (P<0.01) Compared with model group, Changkang tablet group,Qinxiang middle dose group,Qinxiang high-dose group can significantly increase the serum IFN-y levels (P<0.05), Qinxiang low-dose group was not significantly different (P>0.05). the Qinxiang high-dose and middle-dose group's capacity of improving serum IFN-y was significantly better than ribavirin and Changkang tablet group (P<0.05).⑤Serum IL-2 level: Compared with normal group, all built modules were significantly higher (P<0.01); Compared with model group, Changkang tablet group, ribavirin group, Qinxiang high-dose group have significant difference (P<0.05), and Qinxiang high-dose group was superior to ribavirin and Changkang tablet group (P<0.01).⑥Cell morphology showed:normal group is normal, the model group of small intestinal mucosal villous injury and inflammatory cell infiltration, intestinal epithelial cells of drug-treated lesions in varying degrees, but lighter than the model group. Comparison between drug-treated groups, Qinxiang high-dose group to reduce the degree of intestinal epithelial cells vacuolization better than ribavirin group and Changkang tablets group.Conclusion:①HRV oral infection can cause HRV diarrhea model②Every Qinxiang granules dose group can improve the general situation of HRV infected suckling mice, to alleviate symptoms of diarrhea, which Qinxiang high-dose is superior to ribavirin and Changkang tablets.③Qinxiang granules can inhibit virus antigen of HRV infected suckling mice.④Qinxiang granules can enhance serum IFN-γ, IL-2 expression of HRV infected suckling mice and showed a definite dose-effect relationship, which may be part of ways that Qinxiang granules treatment of HRV diarrhea.⑤Qinxiang granules can reduce the small-intestinal mucosal injury and inflammatory cell infiltration, may be its one of the treatment mechanisms of HRV diarrhea.
