Objective To explore the effects of Macrophage Inflammatory Protein-3α/CCL20 Production by Interleukin-17 and Tumor Necrosis Factor-a in Nasal Polyp epithelial cells.Methods Epithelial cells lines,which were established from nasal polyps, were stimulated by culture medium, IL-17,TNF-α.The amount of MIP-3α/CCL20 in the supernatants was measured by enzyme-linked immunosorbent assay (ELISA). MIP-3α/CCL20 mRNA expression was evaluated by reverse transcription-polymerase chain reaction (RT-PCR).Results After stimulated by IL-17 or TNF-α,the expression levels of MIP-3α/CCL20 in nasal polyps were significantly increased compared with those only in culture medium. MIP-3α/CCL20 production synergistically induced by IL-17 and TNF-αwas more than that singly induced.The change of MIP-3α/CCL20 production was in a dose- and time-dependent manner. Moreover,the relative production of MIP--3α/CCL20 mRNA could be measured and got the same result.Conclusions Nasal polyp epithelial cells producing MIP-3α/CCL20,which can be induced by Interleukin-17 and Tumor Necrosis Factor-α,may play an important role in the recruitment of T cells and DCs in nasal polyps.
