The Association Between Aflatoxin Exposure And 8-OHdG And PARP-1 Levels In Liver Tissues From Three Regions Of Guangxi, China

Objective: To study the pathogenesis of Hepatocellular carcinoma, we investigated the levels of 8-OHdG and PARP-1 expression in liver tissues from three regions in Guangxi with varied aflatoxins exposure levels.Materials and methods: 404 liver pathogenesis paraffin wax samples from residents of the three areas were categorized into Nanning group (79 cases) and its control group (58 cases), Liuzhou group (89 cases) and its control group (55cases), Guilin group (86 cases) and its control group (37 cases). Immunohistochemistry method was applied to test liver tissues'8-OHdG level and PARP-1expression of these groups. Correlate analysis method were adopted to observe the existence of the connections between AFB1-DNA and 8-OHdG level, PARP-1expression.Results:â‘ The positive rate of liver tissues'8-OHdG was tested to be 56.6â„…(Nanning group), 51.7%(Liuzhou group),48.8%(Guilin group) in the HCC group. The positive rate of liver tissues'8-OHdG was tested to be 55.2%(Nanning group), 61.7%(Liuzhou group), 48.8%(Guilin group) in the non HCC group. However, the positive rate of the HCC group and the non HCC group were 50.4% and 57.3% respectively, which showed little difference and did not reach statistical significance(P>0.05).â‘¡The liver tissues'PARP-1 expression level was tested to be 75.2â„… in the HCC group and 50.0â„… in the non HCC group. The liver tissues'PARP-1 expression in Nanning group, Liuzhou group and Guilin group were tested to be 89.9%, 76.4%, 60.5% respectively, which has a declining tendency, and was statistically significant (P<0.001). However, in the non HCC group, the liver tissues'PARP-1 expression showed little distinction (P>0.05).â‘¢There is relevant connections between liver tissues'PARP-1 expression and AFB1.Conclusions: The difference in 8-OHdG level among Nanning, Liuzhou and Guilin showed insignificant difference. The PARP-1 expression level of HCC was significantly higher than non HCC. Moreover, liver tissues'PARP-1 level of the HCC group's Nanning group, Liuzhou group and Guilin group has a declining tendency, while in the non HCC group, liver tissues'PARP-1 level of the three locations showed little distinction. We still can't be sure whether there exist any connection between aflatoxins exposure and 8-OHdG level by far. However, HCC's PARP-1 level and AFB1-DNA level have correlativity.
