Objective: The aim is to investigate the effect of poly(ADP-ribose)polymerase(PARP) inhibition on the adherence of platelets to mouse colon carcinoma CT26 cell line in vitro,and how it plays its role.Methods: 5-AIQ served as PARP inhibitior, the effect of PARP inhibiton on the adherence of mouse colon carcinoma CT26 cell to platelets was observed by Tumor cell- Platelets adhesion assay.The effect of PARP inhibiton on the expression of PARP mRNA was detected by RT-PCR in CT26 cell; The expressions of P-selectin and ICAM-1 were investigated by Western Blot in CT26 cell .Results: 1. The adhering platelets of 5-AIQ-treated groups was reduced, when compared with 5-AIQ-untreated group(P<0.05) in the adhesion assay. The inhibitory rate of 5-AIQ group was 28.07%.2. The expressions of PARP and P-selectin and ICAM-1 were attenuated in 5-AIQ-treated group than that in 5-AIQ-untreated group (P<0.05).Conclusion:1. The adhering platelets of 5-AIQ-treated group was reduced, when compared with 5-AIQ-untreated group in the adhesion assay.The result suggests that PARP inhibition can inhibit the adherence of platelets to colon carcinoma CT26 cell in vitro2. The expressions of PARP and P-selectin and ICAM-1 were reduced in 5-AIQ-treated group. The result suggests that the adherence of platelets to CT26 cell was reduced by PARP inhibition in vitro is probably through PARP inhibition, then inhibiting the expressions of P-selectin and ICAM-1. PARP may play a role in colon carcinoma metastasis.
