Objective:To investigate concentrations of 8-hydroxy-2′- deoxyguanine (8-OHdG) and associated molecules in Serum and Urine of nasopharyngeal carcinoma (NPC), head-neck inflammation and healthy controls ,and explore its roles in the tumorigenesis of nasopharyngeal carcinoma(NPC).Methods:ELISA quantitative analysis was used to examine the serum and Urine levels of 8-OHdG and associated molecules(including EB-VCA-IgA,EB-EA-IgG,hOGG1)in sixty-seven cases of nasopharyngeal carcinoma and twenty-eight cases of head-neck inflammation and thirty cases of healthy controls. The relationship between 8-OHdG and associated molecules were statistically analyzed.Results: 8-OHdG level in serum of nasopharyngeal carcinoma group was higher than that of head-neck inflammation group and healthy controls group(P<0.01).But 8-OHdG level in urine , there was no statistical significance among the three groups(P>0.05);In addition, Serum and Urine levels of 8-OHdG in nasopharyngeal carcinoma patients was not related to sex and age(P﹥0.05). hOGG1 level in serum of nasopharyngeal carcinoma group was lower than that of the head-neck inflammation group and healthy controls group(P<0.05 ); EB-VCA-IgA and EB-EA-IgG positive ratio in serum of nasopharyngeal carcinoma group was higher than the other two groups(P<0.05);In nasopharyngeal carcinoma patients, no statistical significance was observed in the concentration of 8-OHdG in serum and urine between the EB-VCA-IgA positive and the negative group(P>0.05). No statistical significance was observed in the concentration of 8-OHdG levels in serum and urine between the EB-EA-IgG positive and the negative group(P>0.05).Conclusion: The increase of 8-OHdG level and the down-regulation of hOGG1 were observed in the serum of nasopharyngeal carcinoma ,suggesting that oxidative damage may be a frequent event in NPC, in which DNA repair capacity may be impaired.In NPC, EB-VCA-IgA and EB-EA-IgG levels were not related to the level of 8-OHdG in Serum and Urine .
