Objective1.To investigate the feasibility of establishing a VX2 breast carcinoma model in rabbit by injection of tumor tissue pieces suspension using percutaneous puncture guided by ultrasound.2.To evaluate the efficacy of new water cooled microwave antenna ablation efficacy for VX2 breast carcinoma in rabbit by means of comparing the result of therapy for VX2 breast carcinoma in rabbit by microwave ablation (MWA) with that of therapy by surgical excision.Methods1.A total of 22 New Zealand white rabbits were used to establish VX2 breast carcinoma models in rabbit by injection of tumor tissue pieces suspension using percutaneous puncture guided by high-frequency ultrasound. The tumor growth was observed with the unaided eye and by high-frequency color Doppler ultrasound. The change of tumor volume was measured and recorded. Three weeks later, use high-frequency color Doppler ultrasound, routine scan of spiral CT and histopathology to indicate the success rate of tumor inoculated.2.25 VX2 breast carcinoma models in rabbit who were 3 weeks established and randomly divided into three groups(A,B,C). The A group was treated by MWA therapy under ultrasound guidance at 25 watts for 120 seconds(10). The B group was treated by surgical excision of breast tumor(10). The C group was control group(5). Observe the imaging change of each group of rabbits using high-frequency color Doppler ultrasound, routine scan of spiral CT.3.All animals of three groups were abserved for three months, then kill the survival animals. The survival rates of all animals and the relapse of tumors in A and B groups were observed. All rabbits were dissected and subjected to physiological analysis after they died. Compare the metastasis of organs of rabbits in each group.Results1.The success rate of vaccination of injection of tumor tissue pieces suspension using percutaneous puncture guided by high-frequency color Doppler ultrasound is 90.9%(20/22).20 laboratory rabbits formed palpable solid tumor after they were planted with tumor tissue pieces one week later, the diameter of the tumor is 0.55±0.11cm. The diameter of the tumor is 1.53±0.16cm.two weeks later and 2.34±0.23cm three weeks later. The size of the tumor increased most after two to three weeks. Then the grow speed was decelerated gradually and irregularly 2.The survival rate of three months of A and B groups were 100% and 90% respectively, they were all significant higher the C group.The metastasis rate of tumor of A group and B group were 25% and 30% respectively in the observing period-three months. Both of them were lower than that of the C group 100%.3.The pathological examination of the implanted metastases, the distant tumor tissue and the metastasis of organs showed they all conformed to squamous cell carcinoma (SCC) of the breast.Conclusion1.VX2 breast carcinoma models in rabbit established by injection of tumor tissue pieces suspension using percutaneous puncture guided by high-frequency color Doppler ultrasound have the following features:easy for operation, having a high success ratio, low cost, avoiding multi-injury to animals, tumor proliferating rapidly, having steady biomedical properties. Their biological behavior is similar to metastatic breast cancer of human beings. They provide ideal animal models for the research and therapy of breast carcinoma.2.MWA can treat VX2 breast carcinoma in rabbit effectively, increase the survival rate of the animals and decrease the metastatic rate of the tumor, which is similar to the curative effect of surgical resection and provide an experiment basis for the clinical research of breast carcinoma treated with MWA.3.High-frequency color Doppler ultrasound and spiral CT are both of great value during the whole period of establishing models and treatment.
