OBJECTIVE Using interphase fluorescence in situ hybridization(FISH) to detect molecular cytogenetic aberrations and explore the characteristics and prognostic significance of molecular cytogenetic aberrations in Chinese patients with chronic lymphocytic leukemia (CLL).METHODS Collecting the bone marrow of 21 patients with chronic lymphocytic leukemia, and of 10 health adults for control ,then using a panel of probes (GLP D13S25,GLP RB1,GLP p53,GLP ATM, CSP12) to detect the cytogenetic abnormalities in 10 health adults and 21 patients with chronic lymphocytic leukemia by interphase fluorescence in situ hybridization (FISH). Cytogenetic aberrations and their relation with some other prognostic factors (sex, age, Binet stage) were analyzed.RESULTS Out of the 21 CLL patients, 15 (71.4%) were found with molecular cytogenetic aberrations. Del(13q14.3) was found in 10 patients(47.6%), del(13q14) in 3 patients(14.2%), at the same time these 3 patients were found to have abnormal del13q14.3. trisomy 12 in 5 patients (23.8%), del(17p13)in 3 patients(14.2%),del(11q22.3)in 2 patients(9.5%).7 patients (33%) showed more than two kinds of abnormalities. Del (13q14.3) was the most frequent abnormality detected, followed by trisomy 12.In these 10 del(13q14.3) patients, more than 60 years ( including 60-year-old)in 6 patients, less than 60 years in 4 patients,male in 5 patients,female in 5 patients, Binet A in 5 patients, Binet B in 3 patients, Binet C in 2 patients.There was no significant relationship between del (13q14.3) and sex,age,Binet stages (both P > 0.05).CONCLUSION CLL has a variety of cytogenetic aberrations,the part has the complex chromosomal abnormalities. Del (13q14) is the most frequent abnormality in CLL. Molecular cytogenetic aberrations detected with FISH have important significance in prognosis evaluation and the choice of therapeutic schedule with CLL.
