ObjectiveTo study on the anti-colorectal cancer effect of Dihydroartemisinin and further explore the mechanism of action.Methods1. In order to understand the current situation of research on colorectal cancer and Dihydroartemisinin, we look up and analyze a large number of domestic and foreign pertinent literatures.2. Makes the molds and the groupings:The bare mouse model building, refers to Zheng Chaoxus'methods. Divides the mouse into 5 groups, each group of 10. Gives the following medicine separately:Normal control group:Fills stomach physiological saline 0.2ml, qd, continual 14d; 5-FU group:Injects into Abdominal cavity 5-FU20mg/kg, qd, altogether 5d:Later will fill stomach physiological saline 0.2ml, qd, until 14d; DHA low dosage group:Fills stomach DHA low dose 30mg/kg, qd, continual 14d; DHA medium dosage group:Fills in stomach DHA dosage 60mg/kg, qd, continual 14d; DHA high dosage group:Fills stomach DHA high dose 120mg/kg, qd, continual 14d.The methods of inoculating nude mice with humancolorectal cancer LoVo which have been well cultivated are used to establish the model of transplanted tumor in nude mice. After that, we divide the nude mouse into five groups and each groups of ten. To intragastric administed Dihydroartemisinin according the low dose, middle dose and high dose (30mg/kg,60 mg/kg,120mg/kg) for 14 days. The tumor suppression effects of Dihydroartemisinin were studied from the following aspects:the xenograft of human colorectal cancer were excised and weighted to calculate the tumor growth inhibitory rate.3. To prepare the pathological section of the xenograft of human colorec of SABC immunohistochemical methods to detect the expression of proteinic factor survivin and caspase-3. in tumor-bearing nude mice.Result1. The result of the expression of proteinic factor survivin shows that the control group's positive rate is 59.30%, the group of 5-Fu is 45.81% and the group of the Dihydroartemisinin high dose is 32.60%. The Dihydroartemisinin high dose group which compared with the control group in difference has statistical significance(P<0.01).2. The result of the expression of proteinic factor caspase-3 shows that the control group's positive rate is10.46%, the group of 5-Fu is 16.74% and the group of the Dihydroartemisinin high dose is 22.15%. The Dihydroartemisinin high dose group which compared with the control group in difference has statistical significance (P<0.01).3. The tumor growth inhibitory rate of the Dihydroartemisinin high dose is 36.54%, the tumor growth inhibitory rate of the Dihydroartemisinin middle dose is 26.55%, the tumor growth inhibitory rate of the Dihydroartemisinin low dose is 12.43%. This inhibitory action shows the dose-effect dependent. The average tumor weight of the high dose group which compared with the control group in difference has statistical significance(P<0.05).ConclusionDihydroartemisinin shows its anti-colorectal cancer effect by influencing the expression of the anti-apoptosis factor survivin and by influencing the expression of the apoptosis factor caspase-3.
