Object : Observe the expression of Survivin,Caspase-9 and Ki-67 in Colorectal cancer,Colorectal adenoma and normal colorectal tissues,analysis the relationship between the expression of three genes in colorectal carcinoma and the biological behavior of Colorectal cancer.By analyse the relationship among them and the correlation with clinical pathological features,research their roles in the development and progression of colorectal cancer,To explore the significance of diagnosis and treatment of Colorectal cancer,and to provide a theoretical basis for early diagnosis and treatment for Colorectal cancer.Methods:Using EliVision Immunohistochemical method to detect the localization and expression of Survivin,Caspase-9 and Ki-67 in 50 cases of Colorectal cancer,30 cases of Colorectal adenoma and 30 cases of adjacent colorectal normal tissues.The expression of Survivin,Caspase-9 and Ki-67 in Colorectal cancer,Colorectal adenoma and normal colorectal tissues which every 20 cases were detected by using Western-blot technology in addition.Results:Immunohistochemistry showe that the positive rate of Survivin in Colorectal carcinoma,Colorectal adenoma and normal colorectal tissues are 78%,50%,20%,there have statistical difference among the three groups.The positive rate of Caspase-9 in Colorectal carcinoma,Colorectal adenoma and normal colorectal tissues are 32%,56.67%,83.33%,there have statistical difference among the three groups.The positive rate of Ki-67 in Colorectal carcinoma,Colorectal adenoma,Colorectal normal tissues are 72%,26.67%,13.33%,the expression of Ki-67 in Colorectal cancer is significantly higher than that in adenoma and normal tissue.However,there is no significant difference in the positive rate of Ki-67 in adenoma and normal colorectal tissues.In Colorectal cancer,the expression of Survivin and Caspase-9 is negatively correlated with the expression of Ki-67,and the expression of Caspase-9 is negatively correlated with the expression of Ki-67.There are no statistical signification between the expression of Survivin,Caspase-9,Ki-67 in colorectal cancer and the patients' gender,age,tumor location,tumor size and depth of tumor invasion.But the degree of tumor differentiation,lymph node metastasis and Duke stage correlated.The results of Western-blot show that the expression of Survivin inColorectal cancer is higher than that in Colorectal adenoma and normal tissues,but in Colorectal adenoma and normal tissues,the expression compared with no significant difference.The expression of Caspase-9 in Colorectal cancer is lower than its expression in Colorectal tumor,lower than the expression in normal tissues,the three groups have significant difference compared with each other.The expression of Ki-67 in Colorectal cancer is higher than expression in Colorectal adenoma,higher than the expression in normal tissues.The three groups are compared with each other with statistically significant difference.Conclusion : 1.The overexpression of Survivin and Ki-67 protein,and the low expression of Caspase-9 protein in Colorectal cancer may lead to abnormal cell proliferation and apoptosis,which may play an important role in the occurrence and development of Colorectal cancer.2.The decreased expression of Caspase-9 and increased expression of Survivin,Ki-67 are significantly related to the malignant biological behavior index of colorectal cancer including tumor differentiation,lymph node metastasis and Dukes stage.These three may play a synergistic role in the occurrence and development of Colorectal cancer,suggesting that they could be used to judge the biological behavior of Colorectal cancer 3.During the evolution of Colorectal cancer,Survivin may inhibit the expression of Caspase-9,make the cell apoptosis cannot be effectively happened,then the cell proliferate abnormally,so as to promote the occurrence and development of Colorectal cancer,There are positive correlation between Survivin and Ki-67 which reflect high proliferation rate of Colorectal cancer.The expression level of Caspase-9 could reflect the colorectal cells ' benign and malignant degree,and its activation could promote the apoptosis of Colorectal cancer cells,inhibit the proliferation of cancer cells,Caspase-9 is negatively correlated with the expression of Ki-67.The differential expression of three tumor associated factors in Colorectal cancer is significant for study the occurrence and development of Colorectal cancer.Combined detection is helpful for early diagnosis and prognosis evaluation of colorectal cancer,and has potential application value in gene targeted therapy for Colorectal cancer.
