Ovarian tumors (ovarian cancer, OC) is one of the three malicious tumors of femals genital tract,the mortality rate ranks top in gynecological malignancieses, because of its relatively hidden nature, many of ovarian cancer are already at a late stage when diagnosed. In ovarian cancer, the epithelial ovarian cancer (epithelial ovarian cancer, EOC)claims about 90% overall, Cytoreductive surgery and platinum-based chemotherapy in epithelial ovarian cancer are the main principles of treatment. However, the following drug resistance of tumor cells causes a considerable number patients seeing further progression or recurrence of disease after chemotherapy, resulting in the 5-year survival rate of less than 30%. Therefore,seeking for indicators of early diagnosis of malignant tumors and to find targeted therapy of gene has gradually become a research direction. Osteopontin (osteopontin, OPN) is a recent discovery of a phosphorylated glycoprotein, and its closely related to cancer development and metastasis. Recent studies have shown that matrix metalloproteinase-9 (matrix metalloproteinases-9, MMP-9) plays an important role in tumor invasion and metastasis process. However the relationship between the two are rarely researched. Early diagnosis and treatment of ovarian cancer has been a hot topic in gynecology research, this study used immunohistochemical methods to determine biological markers of OPN and MMP-9 gene under various physiological and pathological conditions in the ovarian tissue, hence providing theoretical basis to take effective preventive measures, select new therapeutic targets and produce effective treatment programs.Methods Immunohistochemical staining S-P methods was used to detect the expression of OPN and MMP-9 in 60 cases epithelial ovarian carcinoma,20 cases benign ovarian tissue,20 cases borderline ovarian tumor and 10 cases of normal ovarian tissue. every batch of staining are compared with HCC samples for positive-control, and replacing parmingen with phosphate solution as negative control SPSS16.0 software was employed to analyze all data,the data factors with Indepent sample Dunnett T-test and Anlysis of variance;And the relationship of the two factors was analyze by spearman rand correlation test.p<0.05 with significance.Results1. The expression of OPN and MMP-9 in epithelial ovarian carcinomawere significantly higher than those in borderline ovarian tumor, benign ovarian tissueand normal ovarian tissue,The expression of OPN and MMP-9 have relationship with tumor grade(Well Differentiated vs Moderately Differentiated, P<0.05;Moderately Differentiated vs Pooly Differentiated, P<0.05),tumor stage(P<0.05)and Lymph node metastasis(P<0.05), while have no relationship with histological grading(P>0.05)2. There was significant positive correlation between OPN and MMP-9 in epithelial ovarian carcinoma(r=0.80, P<0.05)Conclusion1. The expression of OPN and MMP-9 have relationship with tumor grade,tumor stage and Lymph node metastasis,while have no relationship with histological grading. The increased expression of OPN and MMP-9 may play an important role in the process of carcinogenesis, progression and differentiation of epithelial ovarian carcinoma,these may be used to determine the biological behavior of EOC and indicator of prognosis.2. There was significant positive correlation between OPN and MMP-9 in epithelial ovarian carcinoma, these may be cooperation in the process of carcinogenesis, progression and differentiation of EOC.
