Objective:To investigate the matrix metalloproteinase-9 (MMP-9) polymorphism in Zhejiang Han nationality and study the relationships of this genetic variation with Coronary heart disease (CHD).Methods:Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) was used for the detection of MMP-9 genotype on 128 CHD patients confirmed by coronary arteriongraphy, including 29 myocardial infarction (MI), 58stable angina pectoris(SAP),41 unstable angina (UA) and 106 normal subjects, and then the frequencies of genotype were statistically computed.Results:Three genotypes of MMP-9 including CC, CT and TT had been detected in both groups. The frequencies of CC, CT and TT genotype in CHD group were 75.78%,23.44% and 0.78% respectively, while 85.85%,4.15%and 0% in normal group. The frequencies of CT+TT genotype in CHD group were24.22%, significantly higher than those in the normal group 14.15%. T allele there was 12.50% in CHD group, significantly higher than those in the normal group 7.08%. When analyzed by subgroup, there was no significant difference between the patients with 1-vessel coronary artery disease (CAD),2-vessel CAD and 3-vessel CAD. Then we divided CHD patients into three groups:stable angina pectoris, unstable angina and myocardial infarction group. Frequencies of MMP-9 three genotypes and the T allele were not significantly different between the three groups.Conclusion:our study demonstrated that the polymorphism of MMP-9 gene CT and TT, and T allele was associated with the risk of CHD. But MMP-9 polymorphism may not be useful as a predictor of the severity of coronary heart disease.
