Objective: To exploreβ-catenin erpresses changes and functions in ultraviolet rays-induced mouse cutaneous tumors.Methods: Seventy BABL/C mouses were divided into five groups randomly:control group,two-weeks irradiated group,four-weeks irradiated group, six-weeks irradiated group and eight-weeks irradiated group.Ten mice were biopsied randomly in each group when they finished the ultraviolet rays radiation and then the pathological changes of cutaneous tissue in different stages were detected by HE staining, the expression ofβ-catenin were detected by immunohistochemitry and Real time PCR.Results: Alone with the ultraviolet rays radiation, exposed skin tissue presented inflammation to tumor pathological change. The abnormal expression rates ofβ-catenin in the irradiated four-week group,the irradiated six-week group and the irradiated eight-week group were higher than that in the control group(P<0.05).There was no significant difference between the irradiated two-week group and in the control group(P>0.05).The expression of ?-catenin in nucleus in four-weeks irradiated group,six-weeks irradiated group and eight-weeks irradiated group were higer than that in control group(P<0.05),but there were significant difference among these three groups(P<0.05), there was no significant difference between in two-weeks irradiated group and in control group(P>0.05). The relative expression of ?-catenin mRNA were dectected by the Real time PCR ,and the results showed ?-catenin mRNA was regularly increased beween control group, two-weeks irradiated group,four-weeks irradiated group,six-weeks irradiated group and eight-weeks irradiated group, and there were significant difference among these five groups(all P<0.05).conclusion: 1. High dose wide spectra UVA and UVB light can induce cutaneous squamous cell carcinoma in BABL/C mouse in a short time, and the pathological changes was similar to human's SCC. 2. The expression of ?-catenin translocate into nuclear in ultraviolet rays-induced SCC . 3. We think the abnormal activation of Wnt/β-catenin signal pathway may take part in the process of skin stem cells changing into tumor stem cells , skin tumors are formed.
