Objective To investigate the expression and role of IGF2 and IGF2R in different phases of infantile hemangiomas and vascular malformation.Methods 13 samples from proliferating phase of infantile hemangiomas,10 samples from involuting phase of infantile hemangiomas,15 in vascular malformation and 15 in normal skin samples were involved inthis study. The expression of IGF2 and IGF2R were examined in different in vascular malformation and 15 in normal skin samples were involved in phases of infantile hemangiomas and vascular malformation and normal skin using SP method of immunohistochemieal technique.An image analysis system (Image—pro plus 6.O)was used to measure the average integrated optical density and the rate of positive area of IGF2 and IGF2R in infantile hemangiomas and vascular malformation and normal skin.Results In the proliferating phase of infantile hemangiomas, a number of vascular endothelial cells highly expressed IGF2 and IGF2R.In the involuting phase of infantile hemangiomas, most of vascular endothelial cells or scattered vascular endothelial cells expressed IGF2 and IGF2R.All the samples of vascular malformation and normal skin had no expression of IGF2 and IGF2R.The expression of IGF2 and IGF2R was significantly higher in different phases of infantile hemangiomas than vascular malformation and normal skin(P < 0.01). There was also difference between proliferating and involuting phase of infantile hemangiomas(P<0.01). Conclusion IGF2 and IGF2R maybe play a very important role in the development of infantile hemangiomas. Furthermore, there was another possibility that IGF2 and IGF2 had became a relatively specific factors in involuting phase of infantile hemangiomas. Their significantly different expressions in positive samples,to a certain extent,are related to the stages of infantile hemangiomas.
