Objective To explore the mRNA expression of insulin—like growth factor-Ⅰ(IGF-I), extracellular signal regulated kinase(ERK),glucose transporter4(GLUT4) and insulin receptor substrate-1(IRS-1) in greater omental adipose tissue of metabolic syndrome with colorectal cancer, and to explore the relationship with colorectal cancer. Methods Totally65patients given opportunity-selected operation in Department of Obstetrics and Gynecology and Department of General Surgery, Affiliated Hospital of Qingdao University Medical College, were enrolled and all agreed the content. They were divided into two groups according to the diagnostic criteria of MS:①MS group (n=35), which was subdivided into two subgroups:colorectal cancer (n=15) and non-colorectal cancer (n=20).②Control group(n=30). The fasting blood was taken on the day before operation to determine the fasting blood-glucose and fasting insulin. About200mg greater adipose tissue was removed during operation for RNA extraction. The mRNA expression level of IGF-Ⅰ、ERK、GLUT4、IRS-1in greater omental adipose tissue of the subjects was measured by one-step semi-quantitative RT-PCR,and explore the relation between them. Results①The mRNA expression level of IGF-I and ERK in the MS group was significantly higher than the control group (P<0.01).The level of GLUT4was obvious lower(P<0.01).The expression level of IGF-I and ERK in colorectal cancer group was significantly higher than the non-colorectal cancer group.There was no significant difference between the two groups in the level of IRS-1.②In the two groups,the mRNA expression level of ERK was positively correlated with that of IGF-1(r=0.608,P<0.01).③The fasting insulin was positively correlated with that of ERK and IGF-1, respectively (r=0.538,0.439, P<0.01),and negtively with that of GLUT4(r=-0.457, P<0.01). Conclusion The mRNA expression of ERK and IGF-1in metabolic syndrome with colorectal cancer patients increases significantly. There may be relationship between ERK and IGF-1and metabolic syndrome with colorectal cancer. The GLUT4degree can be one of molecular mechanism in insulin resistance of metabolic syndrome.the relation of IRS-1with metabolic syndrome should be a further research.
