Objective To observe the roliferation, differentiation and IGF-1mRNA expression of extraocular muscle satellite cells in medial rectus muscle of cat model with strabismus, and to investigate the possibility of drug treatment concomitant exotropia。Methods We selected9little cats, which were in sensitive period (4-6w). All animals were randomly divided into3groups: experiment medicate group (group A), experiment control group (group B) and common control group (group C). Each group has3little cats. Group A and group B were made exotropia model by surgical methods. Collected medial rectus muscles of each group three months after strabismus surgery and culture in vitro. Group A was given100ng/ml exogenous IGF-1, group B without it. Compare the changes of the growth curve and rate of myotube formation of each group. Give three groups detection of RT-PCR, Compare the changes of IGF-1mRNA expression through three groups.Results①Growth curve of satellite cells: the curve of group A was higher than that of group B, and the peak of group A shift left; the curve of group C was higher than that of group B,and the peak of group C shift left, too; there were no differences between group A and group C.②Rate of myotube formation: the rate of group A and group C were both higher than that of group B, and the time to achieve maximum integration were earlier than that of group B; There were no differences between group A and group C.③the expression of IGF-1mRNA: IGF-1mRNA expression of group A was higher than that of group B, there was obviously statistical significance between the two situations(P<0.05), and the IGF-1mRNA expression of group C was higher than that of group B, there was obviously statistical significance between the two situations(P<0.05). There were no statistical significance between group A and group C (P>0.05)Conclusion①The speed of the roliferation and differentiation of extraocular muscle satellite cells of experimental strabismus cat model was more deceleration than that of the common cats.②The expression of the IGF-1in extraocular muscles in experimental strabismus cat model was more reduction than that of the common cats.③Exogenous IGF-1may enhance the speed of the roliferation and differentiation of extraocular muscle satellite cells of experimental strabismus cat model.
