Study On Dang Gui Minimally Invasive Acupoint Implants Affect On Shen Xu POF In Mice

Objective:to probe the effect of dang gui minimally invasive acupoint implants therapy on E2 and P in Shen Xu POF (premature ovarian failure, POF) mice caused by tripterygium wilfordii polycoride (TWP) through the treatment principles of. benefiting kidney and spleen, comforting liver and blood. To explore the effect of dang gui minimally invasive acupoint implants in endocrine function of ovarian, and confirm the dang gui minimally invasive acupoint implants'functions in reducing the side effects of ovarian caused by using TWP and investigate it's mechanism.Methods:40 mice were randomly divided into normal control group, model group, western medicine group and dang gui minimally invasive acupoint implants therapy group. The normal control group was intragastric administration by distilled water 40mg/kg per day, continue 10 days. The model group was intragastric administration by TWP 40mg/kg per day, continue 10 days. The western medicine group was established as same as the model group, meanwhile, treated with premarine 0.075mg/Kg by intragastric administration. Dang gui minimally invasive acupoint implants therapy group was also established as same as the model group, meanwhile, treated with dang gui acupoint implants in RN4, SP6, ST36, LR3, one time in five days.The spirit, activity, fur, weight were observed. Estrous cycles were observed everyday until 10 days. On the eleven day, all mice were killed by cutting their neck. Weight the ovaries and uterus. The levels of estradiol (E2) and progestin (P) in peripheral blood were measured with enzyme-linked immunoadsorbent assay (ELISA).Results:1. the estrous cycles of model group prolonged to 7-9d. The estrous cycles of western medicine group and dang gui minimally invasive acupoint implants therapy group is as same as normal control group, it's 4-5d.2. Contrasting with normal control group, the mice of the model group have manifested decreased weight, decreased weight of ovaries and uterus and decreased level of E2 (P<0.05); the weight, weight of ovaries and uterus and level of E2 in western medicine group and dang gui minimally invasive acupoint implants therapy group is little decreased by contrasting with normal control group. The difference between them is not obvious (P>0.05). Contrasting with normal control group, all the decreases of P level in model group, western medicine group and dang gui minimally invasive acupoint implants therapy group are little, but not obvious(P>0.05).Conclusions:through benefiting kidney and spleen, comforting liver and blood, dang gui minimally invasive acupoint implants can increase the E2 level of shen xu POF mice. It can release the side effect of using TWP in ovaries.
