Rohdea japonica (Thunb.) Roth.[Orontium japonicum thumb] has the function of clearing away heat and toxic material, strengthening heart and diuresis. Its indications are including sore throat, diphtheria, mumps, sore swollen drugs, mastitis, Snake bite, heart failure, edema, swelling, hemoptysis, hematemesis.The research of this paper originate from the book "Chinese medicine innovation-Pathogenesis of therapy":the medicine experience of Evergreen in heart failure in Yannian pill prescription. The book contains:Evergreen was invalid under the condition of 30g for decoction nor fried dry,it will also stop the heart in systole in used as powder. According to this book, we can get the following hypothesis:On the one hand the material basis of Rohdea japonica (Thunb.) Roth.may most contained in the water-soluble part than in the fat-soluble part, there may be some heat-sensitive material, easy to sublimation/evaporation material, we may get some information of this material basis by compare crude drug and fried dry drug. On the other hand, the Cure mechanism, the strong clinical side effects of powder may disprove the reliability of clinical experience, the cardiac mechanism of Everygreen may be more than one.This paper base on the research hypothesis,set up different analytic methods of Pharmacology and Pharmacodynatic and analyze different sample of Everygreen. The results are as follow:1,Rohdea japonica (Thunb.) Roth.'s component of an integrated pre-test and the establishment of tracking analysisThe system components of chemical pre-test results showed that Rohdea japonica (Thunb.) Roth. contains cardiac glycosides, polysaccharides, alkaloids, organic acids, which are chemical components with a variety of physiological activities and prospects, confirmed that in addition to cardiac e Rohdea japonica (Thunb.) Roth. glycoside addition there are other active substances. In this paper, We have established different parts of Rohdea japonica (Thunb.) Roth. TLC analysis method and HPLC-UV analysis method. By TLC analys in chloroform and ammonia in the presence of chloroform samples dilute bismuth potassium iodide can be the color of the component; TLC analysis showed that the water extracted samples there is no low-polarity and medium polarity dot.That means active ingredient of clinical experience of Rohdea japonica (Thunb.) Roth., including the broth does not contain alkaloids and terpenes low polarity. Detected on the HPLC peak and acid hydrolyzed samples were analyzed, the results show that HPLC-UV analysis method focuses on the cardiac glycosides were analyzed; Acid hydrolysis of samples of the new test have two obvious peaks.
