| The treatment of the insomnia which caused by imbalance between heart yang and kidney yin of jiaotai pill is the breakthrough point to this reasearch. On the base of the research in early days, the jiaotai pill was extracted into water soluble part,ethanol soluble fraction and volatile oil part by the method of decoction, ethanol, supercritical CO2 extraction,and then by applying the sedative hypnotic pharmacological experiments with these different extract parts, to observe the effects of jiaotai pill on independent activity patterns in mice and hypnosis of pentobarbital sodium of subthreshold dose.and at the same time to analyze the chemical composition of the extraction of different parts,then by analyzing the chemical composition and efficacy of relevance to find out the active ingredients,laying foundations for clinical use and further development of jiaotai pill.By using three methods of the decoction, ethanol extraction and supercritical CO2 extraction to cross into jiaotai decoction, ethanol extract, supercritical CO2 fluid extract three kinds of extracts. the SPF KM mice were divided into saline control group, decoction group and ethanol group, supercritical CO2 extraction group at random,each group were divided into low, medium and high dose group respectively. Each group with low dose (0.3125g/kg.d), medium dose (0.625g/kg.d), high dose (1.25g/kg.d) jiaotai pill extract by intragastric administration,and the normal saline control group administration in accordance with the amount of low dose group (0.3125g/kg.d) equivalent to 0.9% saline, to observe the effects of jiaotai pill on independent activity patterns in mice (the number of activities and standing times of mice) and hypnosis of pentobarbital sodium of subthreshold dose(sleep lantency and sleep duration),furthermore analyzing the difference of different extraction methods on sedative hypnotic efficacy. On the basis of above mentioned jiaotai pill different extraction groups, combined with high performance liquid chromatography method to detecte the content of cinnamic aldehyde, cinnamic acid, berberine hydrochloride in different extracts and analyze the difference extraction methods on their content, By analyzing the relationship between the sedative hypnotic efficacy and chemical composition, ultimately proved jiaotai pill sedative and hypnotic active ingredient. Results were as follows:1.The results of jiaotai pill extracts impact on the independent activity patterns and the sleep time of pentobarbital sodium of subthreshold dosage in mice.Compared with the saline group, jiaotai pill extracts could inhibit the spontaneous activity of mice, and have obvious synergies to cooperate with to affect the sleep time of mice. and supercritical CO2 extraction group was better than the decoction group and ethanol group.2.The results of the determination of cinnamic aldehyde, cinnamic acid and berberine hydrochloride in jiaotai pill extracts.To establish an HPLC method for determination of cinnamic aldehyde,cin- namic acid and berberine hydrochloride in jiaotai pill extracts.chromatographic column:ArcusEP-C18,5μm,4.6×150mm; column temperature: room temperature; flow rate:1ml/min.cinnamic aldehyde chromatorgraphic condition mobile phase:acetonitrile :water(30:70);detection wavelength:290nm;cinnamic acid chromatorgraphic condition mobile phase:acetonitrile:wa-ter(0.1%phosphoric acid)40:60;detection wavelength:267nm;berberine hydrochloride chromatorgraphic condition mobile phase:aceto-nitrile:water(0.033mol/L monopotassium phosphate)25:75;detection wavelength :346nm;the results showed that jiaotai pill supercritical CO2 fluid extracti-on of cinnamic aldehyde,cinnamic acid and berberine hydrochloride was higher than Jiaotai pill decoction and ethanol group,additionally jiaotai pill decoction of berberine hydrochloride was higher than that of ethanol group.3. the chemical composition is the material basis for prescription playing its role. Our experiments showed that the supercritical CO2 extraction had the strongest sedative and hypnotic activities of all the extractions.and it revealed that the content of cinnamic aldehyde,cinnamic acid and berberine hydrochloride was highest.Chemical composition was significantly positivelyassociated with efficacy, indicating that berberine,cinnamic acid and cin-namic aldehyde may be jiaotai pill sedative and hypnotic active ingredient.
