The Research Of Compatibility Mechanism Of "Restoring Normal Coordination Between Heart And Kidney" Of Jiaotai Pill

Jiaotai pill,which is composed of coptis and cinnamon in proportion ten to one.Coptis is great bitter in flavor and cold in property,which can disperse the excessive heart fire,thus to lubricate the kidney;cinnamon is pungent in flavor and hot-natured,which can promote the yang and fire,let the fire back to its origin,promote the yangqi in the kidney and the fire of the gate of life.The combination of coptis and cinnamon indicate the insomnia which caused by imbalance between heartyang and kidneyyin,the clinical signs include disturbed and insomnia,dizziness and tinnitus,morbid forgetful,waist soreness and nocturnal emission,burning sensation of five centres,dry mouth and saliva decreased,red tongue and few music,pulsus parvus and frequent pulse,etc.The traditional Chinese medicine deemed that sleep is the result of normal circulation of yingqi and weiqi,and regularity of invert of yin-yang,if this regularity is demolished,the human body appear hypnic imbalance.The neurophysiologic studies already identified that the regularity of sleep-wake is the result of active movement of central nervous system,and it doesn't dependent on the alternation of day and night.Study show that the hypothalamic-pituitary-adrenal axis is close related in sleep,in the earlier period of sleep,the secretory activity of HPA is inhibited and the slow wave sleep is predominate;in the later period of sleep,the secretory activity of HPA is active and the rapid eye movement is predominate.Hypnic state can be improved through regulating HPA,which also can keep the ratio of the ingredients that sleep related.The regulatory factors of HPA not only regulate HPA,but also regulate sleep directly.In the formula of Jiaotai pill,coptis is bitter in flavor and cold in property,which can inhibit the function of HPA,cinnamon is pungent in flavor and hot-natured,which can active the function of HPA,so we have the conclusion that the mechanism of restoring normal coordination between heart and kidney of jiaotai pill maybe related with the feedback and inverse feedback regulation of HPA.We carry out this experiment in the purpose of finding out the relationship in HPA and "restoring normal coordination between heart and kidney" of jiaotai pill.We use PCPA to deprive the sleep of rats,and break the formula to purging heart group(only coptis),invigorating the kidney group(only cinnamon),restoring normal coordination between heart and kidney group(jiaotai pill),then observe the effects of each experimental group to insomnia state and HPA,choose central neurotransmitter such as 5-HT,DA,NE and cytokine such as IL-1,TNF-αto study and research,use HPLC-ECD to detect 5-HT,DA,NE and ELISA to detect IL-1,TNF-α.The findings of this experiment as follows:(1)the effects to the state of insomnia:we found that after the rats peritoneal injected PCPA to cause insomnia,the rats' circadian rhythm were destroyed,and they showed dysphoria day and night,thus couldn't have normal sleep;we found that jiaotai pill could obviously improve the rats' insomnia, which surpass only use coptis or cinnamon.(2)The effects to the neurotransmitters:the content of 5-HT in the hypothalamus of the insomnia rats were significant reduced,but the content of DA,NE were increased,illustrate that 5-HT maybe promote sleep and DA,NE maybe inhibit sleep;we found that jiaotai pill could increase the content of 5-HT in the hypothalamus and cut down the content of DA,NE,so we concluded that jiaotai pill may through regulating the contents of these neurotransmitters thus to treat insomnia.(3)The effects to the cytokine:the contents of IL-1,TNF-αin the insomnia rats were reduced,showed that IL-1,TNF-αcould promote sleep; we found that jiaotai pill could increase the contents of IL-1,TNF-α,thus could promote sleep.Through this experiment,we conclude that the mechanism of "restoring normal coordination between heart and kidney" of jiaotai pill to treat insomnia maybe relate with the regulating function of HPA.coptis is bitter in flavor and cold in property,which can inhibit the function of HPA and increase SWS,cinnamon is pungent in flavor and hot-natured,which can active the function of HPA,but because it' s on]y ten percent of coptis,so can keep the normal ratio of REM.Coptis and cinnamon,which combination of chills and fever,simultaneous application of purging-tonifying therapy,maybe through selectively active and inhibit different cytokine and neurone to treat insomnia.
