Filipendula purpurea Maxim is the perennial herb for the Filipendula of Rosaceae, which grows in the areas of northeast of China,north of Korea,Japan and Far East area of Russian. As a medicine herb, Filipendula purpurea Maxim was widely used to watered and to the treatment of the rheumatism, epilepsy, gout, frostbite, burns and so on. It is also be used in stanch of Gynaecology. At present, there are few reports about its chemical constituents research and the pharmacological activities reaserch. Especially, there is none research about its chemical constituents. So, in order to make use and develop this plant resources better, I will summarized the chemical consituents and pharmacological activities of the relevant reports and literatures of Filipendula purpurea Maxim first of all.Filipendula belongs to Rosaceae, which main includes these species of Filipendula angustiloba, Filipendula glaberrima, Filipendula intermedia, Filipendula kiraishiensis, Filipendula palmata, Filipendula palmata var. glabra, Filipendula palmata var. palmata, Filipendula purpurea, Filipendula ulmaria, Filipendula vestita, Filipendula x intermedia, widely distributed in northeast of China, north of Korea, north of Japan, Mongolia and Siberia and Far East of Russian.As the deepen study of Filipendula, we found a variety of chemical constituents in the genus, including flavonoids, volatile oil composition, lipid compounds, Tannin compounds and heparin. The fruits of Filipendula contains alkaloids and leaf of Filipendula also contains vitamin C. In addition, amino acid,β- sitosterol-3- O -β- D- glucopyranoside and Monotropitin were found in Filipendula. In recent years, as the research widen, there are some achieved positive progress in pharmacological activities of Filipendula, including antioxidation action,antibacterial action,antitumor action,liver-protection action, immunoregulation action, lower blood pressure action, ischemic-myocardium and brain protection action, antithrombotic and anticoagulant action, promoting fibrinolysis action. Filipendula purpurea Maxim used in this experiment was collected in July 2008 in the Songjianghe Town under Changbai Mountains of Jilin province and assayed by Professor Guangshu Wang of Pharmacy college of Jilin University as Filipendula purpurea Maxim, dried and crushed. First of all, respectively, its chemical composition system pre-experiments with 95%ethanol, 75%ethanol and distilled water for extraction. The results showed that: Filipendula purpurea Maxim on part with 95%ethanol for extraction after heated 3h, filtration, may contain the chemical constituents for the flavonoids, polyphenols or tannins, ,as well as sugar or glycosides, especially rich in flavonoids.So, this experiments mainly study the flavonoids in Filipendula purpurea Maxim.Filipendula purpurea Maxim was extracted three times by 95%ethanol in 80℃,respectivelly,1h,2h and 3h, concentrated and solubled in water, filtered, merged the filtrate, concentrated. The solution are eluted in macroporous resin AB-8 by 20%ethanol and 70%ethanol. The elution are detected by TLC after extracted showed most of flavones existed in the part of 20%ethanol and 70%ethanol. The 20%ethanol extraction was 400g of total flavonoids and taken the use of silica gel column by solvent systems of chloroform-methanol-ethyl acetate-water(4:2:4:1~2:2:4:1). We used Sephadex LH-20 column by organic solvents of methanol and got compound 1 and compound 2. We used the method of re-crystallization and got the compound 3. The 70%ethanol extraction was 120g of total flavonoids and taken the use of silica gel column by solvent systems of chloroform-m ethanol-ethyl acetate-water(4:2:4:1~2:2:4:1). Then, we took the elution use of silica gel column by solvent chloroform-methanol-ethyl acetate-water (5:2:4:1) and Sephadex LH-20 by solvent methanol-water(4:1) and got compound 4. At last, the four compounds were identified as quercetin, kaempferol, quercitrin and hyperin by physico-chemical property and spectral analysis methods. These five compounds are all flavonoids, which were first isolated from this plant.To sum up, the resources of Filipendula purpurea Maxim in our country were rich and the value of development is grate. The study on flavonoids of Filipendula purpurea Maxim of this paper, which provided a scientific basis for th development of new, efficient and safe drug.
