Objective: To provide an experimental basis for clinical application of SUNTOUCH on the hemostatic effect and absorbability as well as liver and renal functional damage and antibacterial effect in vitro.Methods: ( 1 ) The rabbit hepatic hemorrhage wound model was established.They are divided randomly into SUNTOUCH group, S-100 positive control group and blank control group which no hemostatic measures group. The bleeding time and bleeding volume were recorded and compared. In addition, the absorption of SUNTOUCH were observed, 3 rabbits each time, on the 1th, 3th, 7th and 10th day after operation. Meanwhile, we observed the absorption and whether the formation of foreign body after subcutaneous implantation SUNTOUCH from two axillary the same day of operation. Wound tissues were obtained and observed by pathological examination. On the 3th and 5th day after operation, blank control group, S-100 positive control group and SUNTOUCH group were observed by electron microscope in Wound tissues, 2 rabbits each time(.2)On the 10th day after operation,we collected carotid blood from rabbits every group, The levels of ALT, AST, r-GT, ALP, BUN, Cr, UA were detected.(3)Antibacterial on Bacillus subtilis, Pseudomonas pyocyanea, staphylococcus aureus, streptococcus pneumoniae, colon bacillus, Klebsiella pneumoniae were obtained in vitro which were vaccinated by plate streaking. The SUNTOUCH of 3cm×3cm was fixed to the middle of the plates,before culture in the incubator. Colonies of each plate were tested and calculated the inhibition rate.Results: (1)In SUNTOUCH group and S-100 positive control group, the bleeding time were significantly shorter than that of blank control group (P<0.01), the bleeding volume were smaller also (P<0.01). There were no significant differences between SUNTOUCH group and S-100 positive control group(P>0.05). On the first day after peration, the absorption degree of SUNTOUCH in the liver surface was about 90%, and a little white membranous substance were seen, on the 3th, 7th and 10th day after operation, the membranous substance were not found. The liver surface was smooth and no foreign body sensation. It could be considered that SUNTOUCH was absorbed completely. SUNTOUCH in subcutaneous, no residue and granuloma on the first day after operation. Liver pathological histology examination showed that changes of SUNTOUCH group and S-100 positive control group were basically the same, and they both showed no significant difference with the blank control group. Results showed that liver cells similar microscopic changes but no significant variation of the phenomenon to each group on the 3th and 5th day after operation. (2)The index of each group were no significant differences through liver and renal function tests(P>0.05).(3)In vitro, The Bacillus subtilis, Pseudomonas pyocyanea and Staphylococcus aureus were obviously inhibited by SUNTOUCH (P <0.05), and the inhibition rates were 32.52%, 27.66%, 31.84%, respectively. There were no effects on Streptococcus pneumoniae, colon bacillus, Klebsiella pneumoniae (P> 0.05) . In addition, the culture dishes with Bacillus subtilis and Staphylococcus aureus had little bacterial growth on the surface of sticking SUNTOUCH. However, the culture dishes with Pseudomonas pyocyanea had some bacterial growth and the inhibition rate was about 50%.Conclusion: SUNTOUCH has significant hemostatic effect on the rabbit hepatic hemorrhage wound. SUNTOUCH can be completely absorbed on the liver surface after 3 days and absorbed in the subcutaneous tissue after 1 day. Damage on Liver tissue and liver cell are not obvious by liver biopsy and the transmission electron microscopy. Meanwhile the indexes are not significant abnormal in the liver and renal function tests. There was no difference between SUNTOUCH and S-100 in the validity and security. In addition, SUNTOUCH has obviously inhibit effects for Bacillus subtilis, Pseudomonas pyocyanea and Staphylococcus aureus.
