Objective: Evaluate the astringency hemostatic effect of extracts of Callicarpa nudiflora. This study will provied a new approach to the treatment of tuberculosis hemoptysis. The in vitro activity of extracts of Callicarpa nudiflora was evaluated against Mycobacterium tuberculosis and if any, its mechanism.Methods:1. The astringency and hemostatic effect of extract of Callicarpa nudiflora in mice: The experimental mice were divided into groups. Groups were given corresponding medicines for7days and then blood was sampled from mice eyes for the clotting time. The inhibitory effect on inflammation caused by dimethyl benzene in mice was observe as well after7days’administration.2. The in vitro antibacterial effect of extract of Callicarpa nudiflora:Mycobacterium tuberculosis in the logarithmic phases were choosed for the Experiment. The effects against Mycobacterium tuberculosis of Callicarpa nudiflora was tested by double dilution method in tubes.3.The antibacterial mechanism of Callicarpa nudiflora: Cell morphology change of Mycobacterium tuberculosis treated by different concentration of Callicarpa nudiflora were observed with electronic microscope Achievements in protein analysis have been acquired by the combination of SDS-PAGE and mass spectrometry techniques.Results:1. The astringency and hemostatic effect of Callicarpa nudiflora in mice: The research found that after7days’ administration, the group of Callicarpa nudiflora has a shorter clotting time and a better convergence function compared to the control group.2. Callicarpa nudiflora showed excellent activity, with minimum inhibitory concentrations (MBCs) of4.00mg/mL, against the tested three isolates.3. After24h of exposure to Callicarpa nudiflora, M.tuberculosis cells were degraded. The cell wall and membrane dissolved partially or in total. The global protein of Mycobacterium tuberculosis have significant changes in compared with the control group. Three protein bands were revealed as significantly changed before and after the Callicarpa nudiflora treatment. Three high reliability protein were identified from the protein bands.Conclusion1. Our results show that Callicarpa nudiflora help to decrease the clotting time and inhibit the reaction of inflammation caused by dimethylbenzene in mice which can affirm the function in treating tuberculosis hemoptysis.2. In summary, we examined the in vitro anti-TB activity of Callicarpa nudiflora. Our results suggest that Callicarpa nudiflora has excellent activity against M. tuberculosis including drug-resistant strains.3. The Callicarpa nudiflora may functioned as a bacteriostat by influencing the expression of some proteins.
