Background and objective:Prostate cancer(PCa) is a common malignant tumor in Genitourinary system of Chinese Male. The incidence of PCa is going up year by year because of change in life style and structure of diet. PCa accounts for approximately 13% of all cancer deaths in America. In clinical, PSA is used for diagnosis of PCa, however, some diseases such as prostatitis and benign prostate hyperplasia (BPH), even breast cancer also can make PSA level rise. In particularly, it is difficult to distinguish PCa from BPH when serum PSA level is at 4~10ng/ml. affected by the subjective and empirical opinion of the pathologist, the Gleason score may be inaccurate. So it is important to find a more specific biomarker to assess the invasion and prognostic of PCa.Recently CD147 is a hot spot in cancer research. Some researches show that CD147 is a new surface adhesion molecule, also named extracellular matrux metalloproteinases inducer, which can stimulate mesenchymocyte around tumor produce matrix metalloproteinases (MMPs). MMPs can degenerate basal membrane and interstitial substance. CD147 also can up-regulate vascular endothelial growth factor (VEGF) in RNA and protein level, which lead to tumor vessel grow up.In this dissertation, we detect the expression of CD147 in PCa, BPH, normal prostate and fetal prostate by Immunohistochemistry S-P method. Assess the significance of CD147 in diagnosis and prognosis of prostate cancer.Materials and methods:101 cases of PCa, 90 cases of BPH, 36 cases of normal prostate and 15 cases of fetal prostate were collected from guangzhou first municipal people's hospital and cell engineering research centre of the forth military medical university. The average age of PCa patients is 73.5 years old. 101 cases of PCa were divided in I, II, III, IV by TNM stage (5, 30, 38, 28 separately). 101 cases of PCa were also divided in adenomatous hyperplasia, well-differentiated, moderately differentiated, poorly differentiated by pathologic grade (2, 12, 42, 45 separately). The average age of BPH patients is 75.6 years old. The average age of normal prostate patients is 35 years old.Statistical analysis: Data were analyzed by SPSS version 16.01. Data were analyzed by rand test, P values <0.05 were considered significant.2. Survival curve was drawn by Kaplan-Meier method.Result:1. The expressions of CD147 in PCa, BPH, normal prostate and fetal prostate are significantly different. 2. The expressions of CD147 in TNM stage, WHO grade and Gleason score are significantly different separately. 3. There is a negative correlation between CD147 positive expression and prognosis of PCa patients.Conclusion:1. The expression of CD147 in PCa is the highest. 2. There are positive correlation between the expression of CD147 and TNM stage, WHO grade and Gleason separately. 3. CD147 is conduced to raise early diagnosis rate of PCa, it is hope to be a biomarker which can reflect malignant degree and assess the prognosis.
