Objectives:To investigate the expression of signal transducers and activators of transcription 3 (STAT3)and CD147 in human prostate cancer(PCa)to explore their relationship with cell differentiation ,PSA and Gleason Scoring.Methods:To understand the differences of STAT3,CD147 expressions among Benign Prostatic Hyperplasia and Prostate Cancer and to seek out the relationships between the various pathological ratings of prostate cancer and STAT3,CD147 as well as the relationships between PSA and Gleason Scoring, researchers stained 15 cases of Benign Prostatic Hyperplasia specimens and 60 cases of prostate cancer specimens .The expression of STAT3 ande CD147 were dectected by means of immunohistochemistry. The principle of immunohistochemical staining: There are some differential immunolo- gical reactions between marked antibodies and antigens in histiocytes. Then, tumor markers on marked antibodies react to developers. Last, we can definite detected antigens'quantity and cytological location by observing chromogenic objects with microscope.Results:1. The positive expressions of STAT3 and CD147 in prostatic tissues are stained from yellow to brown in cytoplasm. Only one positive expressions of STAT3 and only one positive expressions of CD147 in normal prostate and the color is lighter yellow, the positive rate of STAT3 and CD147 in Benign Prostatic Hyperplasia is 40.0℅ and 26.6%,but the positive rate reach to 73.3℅ and 61.7% in prostate cancer , whose color in cytoplasm is deeper. The positive rates of STAT3 and CD147 in PCa was higher than those in BPH ( P<0.05).2. The positive rates of STAT3 and CD147 in Gleason scoring group were higher than those in low Gleason scoring group.3.All of the negative STAT3 and CD147 expressions are distributed in well differential tumors ,the highly positive rate of STAT3 and CD147in poorly differential tumors are 41.6℅ and 37.5℅;70.0% and 80.0% negative expression in the group of PS cell differentiation A<4; 100℅ positive expression in the group of PSA>20, moreover,57.1℅and 57.1%highly positive expression in the group of PSA> 20.The postive rate of STAT3 and CD147 corresponds with the increase of tumor's pathological grading and its PSA and furthermore with deeper color in cytoplasm.4.And using Spearman rank correlation analysis ,There was a positive correlation between the expression of STAT3 and that of CD147(rs′=0.7757,P<0.01).Conclusion:1.The expression of STAT3 and CD147 in prostate cancer is higher than that in Benign Prostatic Hyperplasia.2. The positive rates of STAT3 and CD147 in high Gleason score group were higher than those in low Gleason score group. The positive rates of STAT3 and CD147 in poorly differential tumors were higher than those in well differential tumors.3. The expression of STAT3 and CD147 in prostate cancer present positive correlation with PSA. The expression presents close relation with the carcinogenesis, progression and metastasis of prostate cancer.
