Objective:This topic uses CD147 as a research target,and uses androgen-dependent prostate cancer(ADPC)cells LNCa P to explore the relationship between CD147 and oxidative stress,and clarify the role and mechanism of CD147 in the oxidative stress of prostate cancer cells,discover new functions of CD147,and provide new ideas for the treatment of prostate cancer.Methods:Using different concentrations of hydrogen peroxide(H202)to stimulate LNCa P cells at different time points,real-time PCR(Real-time PCR)technology was used to detect CD147 expression changes;RNAi interference technology was used to establish an LNCa P cell line with low CD147expression,named as a negative control Group(LNCa P/Scramble)cells and CD147 interference group(LNCa P/sh CD147)cells,using RT-q PCR and Western blot technology to verify the expression of CD147.The kit was used to detect the activity of catalase(CAT),superoxide dismutase(SOD),glutathione peroxidase(GSH-PX)and malondialdehyde in LNCa P/sh CD147 and LNCa P/Scramble cells(MDA)content;flow cytometry to detect the changes of intracellular reactive oxygen species(ROS).Different concentrations of H202act on LNCa P/Scramble and LNCa P/sh CD147 cells,CCK-8 method to detect cell growth,flow cytometry to detect cell apoptosis;PI3K/AKT pathway inhibitor LY294002 is added,and Western blot method is used to detect AKT and p-AKT,Nuclear factor E2 related factor(Nrf2)and heme oxygenase-1(HO-1)expression changes.Results:As the concentration of H202increases and the duration of action increases,the expression of CD147 gradually increases.The results of RT-PCR and Western blot showed that compared with the LNCa P/Scramble group,the CD147 expression in the LNCa P/sh CD147 group decreased in gene and protein level,P<0.01.After silencing CD147,the intracellular ROS and MDA content increased(P<0.01),while while the concentration of CAT,the activity of SOD and GSH-PX decreased(P<0.01).Compared with LNCa P/Scramble cells,with the increase of H2O2concentration,the survival rate of LNCa P/sh CD147 cells decreased(P<0.01)and apoptosis increased(P<0.01).Compared with the LNCa P/Scramble group,the expression of p-AKT,Nrf2 and HO-1 decreased in the LNCa P/sh CD147 group(P<0.01).Compared with the LNCa P/sh CD147 group,the expression of p-AKT,Nrf2 and HO-1decreased after treatment with PI3K/AKT signaling pathway inhibitor LY294002(P<0.01).Conclusion:CD147 can inhibit the oxidative stress of prostate cancer cells and reduce cell apoptosis induced by oxidative stress.Its inhibitory mechanism may be related to the PI3K/AKT signaling pathway.
