| Backgroud: The chronic heart failure is the ultimate conjunct chunnel and the important reason of death of all kinds of heart diseases. The incidence and the mortality of the heart failure is higher and higher with the social advancement and the heavy burden of the society and the family. It is showed that the basal mechanism of the chronic heart failure being taking place and developing is the ventricular reconstruction which includes the series of the change of the cardiac muscle in structure, function and token conduced by the mechanism of the cellular and the molecular in the modern research. Many neural incretory factors as the sympathetic nerve system, the RAAS, the angiotensin and so on have the significant function intervening the hypertrophy.It has been showed that many inflammatory factors and infections have participated in the mechanism of the heart failure lately, and the correlative proof has been discovered in the blood and the cardiac muscle.The foundational theory has proved that the inflammatory factors which have been studied mostly is the tumor necrosis factor-α(TNF-α)and the Transforming growth factor-β(TGF-β)can influence the course of the heart failure, and have the close connection of the other system. Thereby the inflammatory factors is possibly the unattached important way, or possibly the portion of the other systems to affect the hypertrophy and the ventricular reconstruction.There are many research and important fruit about the hypertrophy in the traditional Chinese medicine. It is reported that Zhenwu Tang Granule can improve the symptom of the patients of the heart failure with heart and kidney weakness, and can interact the RAAS, the Endothelin system and the Oxygen free radicals. But there is not more research of the traditional Chinese medicine concerning the inflammatory factors, especially in the hypertrophy. So it is the great value in the research and the clinic that studying the interaction among the Zhenwu Tang Granule ,the hypertrophy and the inflammatory factors.Objective:To research the influence of the Zhenwu Tang Granule acting on the left ventricular reconstruction and the inflammatory factors in rats with the chronic pressure overloaded.Methods:The male SD rats were randomly divided into 3 groups:sham operation group(n=8),operation group(n=15)and herbal medicine group(n=15).Myocardial hypertrophy was induced by gradually constricting the abdominal aorta in rats. After 16 weeks, the herbal medicine group was treated with Zhenwu Tang Granule, a Chinese herbal medicine, 12g/Kg in feedstuff for 8 weeks. Then all rats were assessed for progressive changes in left ventricular(LV) structures and functions with high frequency ultrasound at 24 weeks after operation. Cardiac muscle was stained with HE, myocardial collagen was stained with Masson and CVF-T and CVF-NV were analyzed using a computer assisted procedure. The serous concentration of the TNF-α,TGF-β,PGE2,CRP,UA was be calculated. And the cox2 in cardiac muscle was marked by immune histochemistry.Result:There is much damage with the wall thickness, internal diameter,LV-FS ,EF and cardiac index both in the operation group and the herbal medicine group, but it is more serious in the operation group than in the herbal medicine group, where there is statistic differentia. Also there is significant difference with CVF-T, CVF-NV, the serous concentration of the TNF-α,TGF-β,PGE2, between the operation group and herbal medicine group,but there isn't difference in CRP and UA between the two groups. On the other hand, it is showed that the greatly contrast with the pathologic result which including the mark by HE,Masson, also and the immune histochemistry, between the both mentioned group. But there is no difference between the herbal medicine group and sham operation group.Conclusion: Zhenwu Tang Granule has affected the many inflammatory factors of the serum in rats. Zhenwu Tang Granule has restrained the hypertrophy in rats with the chronic pressure overloaded.And the restrained mechanism is maybe through the inflammatory factors,or the inflammatory factor is one of the restrained mechanism.
