Objective: To detect the expression of visfatin in visceral adipose and serum visfatin levels of type 2 diabetic rats, and to explore the relationship between visfatin and diabetic interrelated factors or insulin resistance. Methods: Male rats were fed with a diet enriched with sucrose and fat for 8 weeks, streptozotocin (STZ) was then injected intraperitoneally to induce a model similar to human type 2 diabetes mellitus. The expressions of visfatin mRNA in visceral adiopose of two group rats were determined by real-time RT-PCR, serum visfatin levels were determined by enzyme-linked immunosorbent assay(ELISA). The relationships between visfatin and fasting blood glucose (FBG), fasting insulin(Fins), free fat acid (FFA), triglyceride (TG), total cholesterol (TC), homeostasis model assessment- insulin resistance index (HOMA-IR) or insulin sensitivity index (ISI) were observed. Result: The expression of visfatin mRNA in visceral adipose and the levels of serum visfatin were significantly increased in type 2 diabetic rats versus normal control rats (P<0.05). The levels of serum FFA, TG, Fins and HOMA-IR were significantly increased, ISI were decreased significantly in type 2 diabetic rats versus normal control rats(P<0.05). Pearson correlation analysis illustrated that the serum visfatin level were significantly positively correlated with FBG, FFA and HOMA-IR. and negative correlation with ISI. Conclusion: The expression of visfatin mRNA in visceral adipose and the serum visfatin levels were significantly increased in type 2 diabetic rats. The increasing of visfatin may be related to the metabolism disorder of glycolipids and insulin resistance.
