Objective: Study of the protection of Dendrobium Candidum Wall.Exlindl Drop(DCWD) in radiation-induced mice lymphocytes and spermatogenic cell apoptosis,to explore its role in the protection of radiation injury.Methods: 100 male BALB/c mice were randomly divided into, normal control group, radiation alone group, DCWD low-dose group and DCWD high-dose group. Before the whole-body irradiation with 4.0Gy 6MVX-ray,the BABL/C mice were supplied with DCWD once a day for 15 days, at 6 hours after irradiation the mice were killed. By using the light microscope to observe the spleen,thymus,testis histological changes at the hematoxylin and eosin staining. Observation of spleen lymphocytes, thymocytes, Spermatogenic Cell apoptosis by SEM, and spleen and thymus lymphocyte apoptosis observing rate changes by FCM. Immunohistochemical method to observe the spleen lymphocytes,thymus lymphocytes and spermatogenic cell apoptosis-related protein Bcl-2,Bax and Bcl-2/Bax ratio.Results: (1) Pathological changes of the spleen, thymus and testis tissue: The radiation alone group showed acute radiation injury emerging of the spleen,thymus and testis, which manifested as disturbance of the blood circulation, cell degeneration and comp adaptation. The DCWD groups showed slighter pathological degrees of change and recovered sooner. There was no difference in the DCWD low-dose group and DCWD high-dose group.(2)By using the electron microscope the changes of spleen and thymus lymphocytes: Based on the changes of hematoxylin and eosin staining, the thymuses and spleens of the males were undertaken electron microcopy scanning. Some features of apoptosis in cell morphous and architecture were observed, as the arrowheads. It showed obvious pycnosis in lymphocytes caryons, degeneration in mitochondria and lymphocytes clusters of apoptosis.(3) Changes in the rate of apoptosis of spleen lymphocytes and thymus lymphocytes: by contrast the model, after irradiated the mice lymphocytes apoptosis was significantly increased. But there was no difference between the DCWD low-dose group and DCWD high-dose group.(4)Expression of Bcl-2 and Bax protein in the spleen thymus and testis tissues: Bax expression was stronger in the model group than in the control group, resulting in the ratio of Bcl-2/Bax to noticeably decrease. Bcl-2 protein expression was induced by DCWD treatment, and the ratio of Bcl-2/Bax was increased. However there was no difference between the two DCWD treatment groups.Conclusion: On contrast DCWD treatment has certain protective effects on acute radiation-injury, and its mechanisms may be related to the changes in architecture of spleen and thymus increase in apoptotic cell numbers, improve the expression of Bcl-2 and the Bcl-2/Bax ratio, and so on.
