This empirical study was divided into two parts to study the protective effect ofDendrobium candidum Wall.ex Lindl drop(DCWD) on acute radiation-injuried mice.Part 1. In Vitro Empirical Study of Dendrobium candidum Wall.ex Lindl drop onRegulating Immunocyte Functions in MiceObjective To study the regulating immunocyte functions of the Dendrobiumcandidum Wall.ex Lindl drop(DCWD) in vitro mice cells.Methods DCWD had some different concentrations by being dissolved in culturemedium (1mg/ml,100ug/ml,10ug/ml,1ug/ml). Study their abilities of blastisation oflymphocyte and the phagocytosis of peritoneal macrophage under the differentconcentrations of DCWD by the MTT way. Inspect the contents of IL-2 by spleniclymphocyte under the stimulus of ConA with DCWD and TNF-αby peritonealmacrophage under the stimulus of LPS with DCWD.Results (1) The blastisation of lymphocyte 1. when the concentration of DCWD was100ug/ml or 1mg/ml, it stronged the ability of the blastisation of splenic T lymphocyte.And there was obvious difference between them(P<0.01). But when the concentration waslower, there were no differences between the results (P>0.05). 2. when the concentrationof DCWD was 1mg/ml, it stronged the ability of the blastisation of thymic Tlymphocyte(P<0.01). But there were no differences between the results under the otherdiffemt concentrations of DCWD (P>0.05). (2) The phagocytosis of macrophage, Therewere no diffences on the phagocytosis of macrophage under the different concentrations ofDCWD(P>0.05). (3) The inspection of cytokines, 1.IL-2, when the concentration of DCWDwas 10ug/ml, it increased the content of IL-2 by splenic lymphocyte(P<0.01). But therewere no differences under the other different concentrations of DCWD (P>0.05). 2.TNF-α,when the concentration was 0.1ug/ml, it increased the content of TNF-α(P<0.05). Butwhen it was 1ug/ml, it decreased the content obviously (P<0.01). Compared to the controlgroup, the content was lower when the concentration was 1ug/ml or 100ug/ml. But there was no difference (P>0.05). But when the concentration was high to 1mg/ml, the contentof TNF-αbecame lower obviously (P<0.05).Conclusions DCWD had some regulational functions on the vitro immune cells ofthe mice. It stronged the ability of the blastisation of lymphocyte and regulated thecontents of correlative cytokines.Part 2. In vivo Empirical study on protective effect of Dendrobium candidumWall.exLindl drop on acute radiation-injuried miceObjective To study the protective effect of Dendrobium candidum Wall.exLindldrop on acute radiation-injuried mice and the correlative mechanism.Methods According to the body weight, 6-8 weeks old female BALB/c mice weredivided into normal control group, radiation-injuried group and DCWD groups which weredivided into two groups (4,2g/kg). Before whole-body irradiation with 4.0Gy 6MV X-ray,the BALB/c mice had been supplied with DCWD once every day for 16 days. After beingirradiated, the mice were continued to be given DCWD until they were killed. The ratios ofspleen and thoracic gland to the whole body weight, the DNA contents of bone marrow,the CD4~+/CD8~+ ratios of peripheral blood and splenic cell, the blastisation of lymphocyteand the contents of IL-2 by splenic lymphocyte and TNF-αby peritoneal macrophage wereobserved.Results (1) General state of health, The consciousness was poor, food and activitydecreased in mice irradiated by X-ray, especially in radiation-injuried group but DCWDgroups were better. (2) The ratios of the spleen and thoracic gland to the whole bodyweight, The spleen and thoracic gland obviously shrinked in mice irradiated by X-ray, butthe DCWD groups were much better(P<0.01). (3) The DNA contents of bone marrow,Compared to the control group, the DNA contents of radiation-injuried group decreasedmuch obviouly(P<0.01). But it was higher in each DCWD group (P<0.01), however, thedifference had no statistical significance between them(P>0.05). (4) The index of immunefunction, 1.T lymphocyte subgroups, The ratios of CD4~+/CD8~+ of both peripheral blood and splenic T lymphocyte were much higher in mice with X-ray irradiated, which wasbecause the CD4~+% higher and CD8~+% lower. The CD4~+/CD8~+of DCWD groups weremuch lower(P<0.01), but there was no difference between them (P>0.05). There was nodifference in the index of CD4~+% of peripheral blood between the control group and thehigh dose DCWD group. But there were obvious differences in the indexes of CD4~+% andCD8~+% of the splenic lymphocyte bwtween the radiation-injuried group and DCWDgroups. 2. The blastisation of lymphocyte, The ability of the blastisation of T,Blymphocyte became poorer in mice with X-ray irradiated(P<0.01). They were muchstronger in DCWD groups(P<0.01), but there was no difference between them(P>0.05). (5)The inspection of cytokines, 1.IL-2, The content of IL-2 decreased much obviouly in micewith X-ray irradiated comparing to the control group. It was much higher in DCWDgroups(P<0.01), but there was no difference between them. 2. TNF-α, It became higher inmice with X-ray irradiated, but the difference had no statistical significance between eachgroup (P>0.05).Conclusion DCWD can protect the acute radiation-injuried mice which relateswith protecting the hematopoiesis and the immune function, and so on.
