The Chinese medicine dictionary says of Dendrobium officinale "designed nourishing lung and stomach of gas liquid, gas-liquid Chong Mong, kidney water spontaneous, there are a lot of Dendrobium candidum nourishing the stomach yin aspects on achievements in scientific research at present, and pharmacological study of nourishing kidney Yin is rarely involved. In pa-tients with kidney-yin deficiency showed low immunity or disorder and reduce the antioxidant capacity, Liuwei Dihuang pill is a recognized treatment of kidney yin deficiency syndrome of classical prescription, clinical and experimental verification of Liuwei Dihuang Pill regulating kidney yin deficiency immune disorders and improve the antioxidant capacity of. So this experiment from the point of view of antioxidant and immune, Liuwei Dihuang pill as a positive control group, exposing different doses of Dendrobium candidum polysaccharide on rat kidney yin vacuity models of pathological effect can not only rich theory to physiology and pathology of the kidney yin deficiency syndrome, and to provide theoretical and experimental basis for the elucidation of Dendrobium officinale polysaccharide of kidney yin deficiency of pharmacological mechanism.MethodThe 50 SD rats were two months old, by pouring gastric hydrocortisone (modeling for six days) Kidney Pathological Model of rat was established. Experimental rats were randomly divided into model group and blank control group (saline group), treatment group (Dendrobium officinale polysaccharide high dose group, Dendrobium polysaccharide low dose group and positive control group (Liuwei Dihuang pills group), after the end of the experiment, detected the serum of anti oxidative and immune indices of content, and carries on the comparison between groups, the verification of Dendrobium officinale polysaccharide of kidney yin deficiency rats induced by pharm-acological mechanism. The antioxidant index set for cortisol (CORT), malon-dialdehyde (MDA), superoxide dismutase (SOD); immune index set for immunog-lobulin M (Ig-M), leukocyte mediated)-6 (IL-6), immunoglobulin G (Ig-G) and leukocyte mediated biotin 2 (IL-2).The experimental results(1) weightThe rats weight before the experiment had no significant difference (P> 0.05), weight have varying degrees of increase in the experimental process, after the end of the experiment, the body weight of the rats in the blank group growth the fastest, Liuwei Dihuang pills rats, the body weight growth relatively slow, kidney yin deficiency model of rats weight grew at the slowest rate, growth of Dendrobium officinale polysaccharide high dose group and low dose group, the weight of the relative experimental group grew fa-ster. Slightly lower body weight of rats significantly decreased the weight of the body weight of the rats after statistical analysis to the medicine after each group and blank group compared with that of the kidney yin defici-ency model group, with significant difference (P<0.01), Dendrobium poly-saccharide high dose group, low dose group than in the control group, but there was no significant difference (P>0.05), Liuwei Dihuang pills group weight loss between groups differences (P<0.05); and kidney yin deficiency model group compared to the Dendrobium officinale polysaccharide low dose group and high dose group and the body weight of rats increased significantly. There were significant differences (P<0.01), Liuwei Dihuang pills group elevated body weight between groups differences (P<0.05).(2) CORT concentration in serum of ratsCompared with normal rats, CORT content in serum of rats in model group increased significantly (P<0.01). Compared with the model group, the Liuwei Dihuang pill, Dendrobium candidum in high dose group, low dose group rats serum CORT content significantly decreased (P<0.01) and high dose of Polysaccharide from Dendrobium reduce kidney yin deficiency rat serum CORT effect is superior to that of Liuwei Dihuang pill, and between groups in serum CORT levels have difference (P<0.05).(3) the concentration of SOD and MDA in serum of ratsCompared with the rats in the normal group, activity levels of SOD in serum of rats in the model group was significantly decreased, MDA content increased significantly, with statistically significant difference (P< 0.01); Liuwei Dihuang pills group, Dendrobium polysaccharide high dose group, low dose group, serum SOD activity were significantly increased (P<0.01), Dendrobium polysaccharide high dose group rats serum MDA content decreased significantly, Liuwei Dihuang Pill group, the serum MDA content decreased significantly (P<0.05), Dendrobium polysaccharide low dose group rats serum MDA content although decreased, but there was no statistical significance (P>0.05)(4) in rat serum IL-2, IL-6, Ig-M, Ig-G concentrationAlthough, Ig-M compared to, Ig-M Shi Huduo and reduce significant content of (IL-2 significantly, Ig-M, Ig-G compared to rats and the normal group, the model group rats serum IL-2 content was significantly decreased (P<0.01), IL-6 levels increased significantly (P<0.01); and the model group compared with Liuwei Dihuang pills group, Dendrobium officinale polysaccharide high dose group, low dose group rats were significantly Xing Shenggao (P<0.01), the Ig-G of the (P) and Liuwei Dihuang Pill sugar high dose group rats serum IL-6 content was significantly decreased (P<0.01), but with the model group, Dendrobium polysaccharide low dose group, IL-6 levels decreased, but the difference was not statistically significant (P>0.05). Conelusion(1) to rats irrigation stomach hydrocortisone can be successfully produ- ced pathological model of kidney yin deficiency rats and showed signs of kidney yin deficiency syndrome change and adrenal cortex hyperfunction (serum CORT significantly increased).(2) reduce the activity of superoxide dismutase (SOD) in serum of rats in the model group, the serum MDA levels were significantly increased, suggesting decreased antioxidative ability of kidney yin deficiency rat. The rats in the model group, the serum IL-2 expression level was significantly reduced, IL-6, Ig-G, Ig-M expression levels were significantly increased and that the kidney yin deficiency rat in a state of immune disorders.(3) high dose of Dendrobium officinale polysaccharide can obviously correct the kidney yin deficiency rat immune disorder state and improve the antioxidant capacity of the rat, with improved pharmacological effects of kidney yin deficiency syndrome in rats and curative effect is better than that of Liuwei Dihuang pill, low doses of Dendrobium officinale polysaccharide effect is not significant.(4) high dose and low dose of Dendrobium officinale polysaccharide can significantly increased the weight of kidney yin deficiency rat, the body weight of the rats in the two groups although slightly lower than the blank group, but between the groups no difference, the curative effect is better than that of six taste glutinous rehmannia pill.
