Objective:â‘´To investigate the improved methods of preparing for animal model of fetal loss induced by APA.⑵To explore the influence of Dan Shou Tang to fetal loss induced by APA.Methods:â‘´10-week-old SD pregnant rats were randomly divided into 5 groups: A, B, C, D and blank control group. On the 8th and 12th day of pregnancy, multi-site subcutaneous injection of ACA or LA of protein solution on the back was used to the every rat of group A in the dosage of 25 mg/(kg·d), B in 50mg/(kg·d),C in 75mg/(kg·d), and D in 100mg/(kg·d). Until the 15th day of pregnancy, they were killed for taking samples. Meanwhile, the serum of healthy human was taken as a blank control group. Absorption rate and weight of fetus were calculated; OD of ACA was measured by enzyme mark instrument and activities of LA determined by activated partial prothrombin time (APTT).⑵10-week-old SD pregnant rats were divided randomly into five groups. The rats in Groupâ… were received the treatment of nourishing kidney and promoting blood flow; the rats in Groupâ…¡were received the treatment of nourishing kidney; the rats in Groupâ…¢were received the treatment of promoting blood flow; Groupâ…£-APA model group and Groupâ…¤-blank group. From the first day of pregnancy, the rats in Groupâ… were given Dan Shou Tang through intragastric administration; the rats in Groupâ…¡were given formula to nourish kidney and rats in Groupâ…¢were given formula to promote blood flow, while the rats of APA model group and blank group, as the control were given the corresponding physiological brine through intragastric administration. Then on the 8th and the 12th day of pregnancy, all rats were given multi-site subcutaneous injecting of purified ACA and LA protein solution with a dosage of 15mg/ml. On the 15th day of pregnancy, the rats were killed for samples. Meanwhile, the serum of healthy human was taken as a blank control group. Absorption rate and weight of fetus were calculated; OD of ACA was measured by enzyme mark instrument and activities of LA determined by APTT.Results:â‘´Studying the rat model, we found that the number of ACA in serum and embryo absorption rate increased significantly (P<0.05), and APTT prolonged, while embryonic weight decreased significantly (P<0.05). The results of pathological observation showed some phenomena such as deposit of placenta immune complex, tissue degeneration and necrosis, inflammatory cell infiltration, fibrin agglutinations and blood stasis in the vessel, etc, which were all positively correlated with dosages of antibody protein solution. In addition, 75 mg/(kg·d) ACA protein solution or LA protein solution is the stable and effective dosage of immune antibody for the model.⑵Compared with APS model group, Groupâ… ,Groupâ…¡and Groupâ…¢can dramatically decrease APA level and fetal absorptivity, and increase fetus weight. Furthermore, the differences are of great significance (P<0.05); Compared with the blank group, Groupâ… had no significant statistic difference in APA level,fetal absorotivity and fetus weight (P>0.05). However, Groupâ…¡and Groupâ…¢had significant differences (P<0.05).Conclusion:â‘´SLE patients serum can be one of the immune antibody sources to replicate the animal model; 75 mg/(kg·d) ACA protein solution or LA protein solution is the stable and effective immune antibody dosage for a model on each time; the improved animal model conforms to the biological, obstetrical and pathological changes of fetus loss patients induced by APA; the improved model is stable and greatly repeatable.⑵The formula with function of nourishing kidney and promoting blood flow has positive clinical efficacy in decreasing APA level,fetal absorptivity and increasing fetal weight and pregnancy chances.
