Experimental Study On The Effect Of Modified Buxindan Decoction On Behavior And Monoamine Neurotransmitters Of Rats With Insomnia

[Objective] Set up the insomnia-model to estimate the treatment of modified Buxindan decoction on insomnia. Investigate the influence of modified Buxindan decoction on insomnia behavior study and the monoamine neurotransmitters, and through it to explore its mechanism of treatment of insomnia of modified Buxindan decoction, and improve Tianwangbuxindan of medication safety factor, which for this drug in future clinical safety of using experimental data to provide scientific.[Methods] The insomnia rat model was set up by the P-chlorophenylalanine (PCPA). By observing modified Buxindan decoction autonomous activity in rats, the effects of pelltobarbitalum natricum in mice and the sleep time induced by pelltobarbitalum natricum hypnosis subliminal impact, examining the efficacy of its treatment of insomnia. Fluorometric method is applied to detect the monoamine neurotransmitters (NE and 5-HT) of the brains. To evaluate the efficiency of modified Buxindan decoction on the insomnia behavior tests and cerebral monoamine neurotransmitters of insomnia rat model through statistics means.[Results] Compared with the blank control group, model of insomnia in rats has a very significant difference in autonomous activity and the monoamine neurotransmitters (NE and 5-HT) of the brains. the high-dose group of modified Buxindan decoction and Tianwang Buxin Boluses showed similar efficacy. Pharmacodynamic experiments showed that there is no significant differences between modified Buxindan decoction and Tianwang Buxin Boluses.The two drugs all can extend the sleep time of the mice, and increase the number of sleep in mice by pentobarbital sodium.[Conclusions] The insomnia rat model by PCPA is the ideal model of insomnia for the reason of relatively simple, short-time, the high successful rate, repeatability, better stability. Tianwang Buxin Boluses without zhusha can still exert a considerable effect of drugs to improve the animals sleep.and the must was the dose-effect relationship, The role of the principle of treatment of insomnia of Buxindan decoction mainly is due to increasing the cerebral NE and 5-HT concentration in the brains.
