Studys On Mechanism And Screening Of Chinese Herbal Medicine Complex Prescriptions To Expedite Wound Healing

The purpose of the experiment was to provide a theoretical basis for clinical veterinary medicine by studying the screening of compound traditional Chinese medicine to promote wound healing and its mechanism.In this study, Wistar rats as experiment animal,they were perforated by self-made hole puncher at the back on both sides to built models of skin defects.Combined with the theory of traditional Chinese veterinary medicine ,on the basic of Fangfushengji powder ,and invigorating blood circulation and eliminating stasis drug , removing necrotic tissue and promoting granulation drug were chose as the main drug, heat-clearing and damp-drying drug and apocrustic astringent were chose as the adjuvant,all of these as the principles of prescription component.We selected dragon's blood,Frankincense, myrrh, Sanchi, Shikonin, Angelicadahurica, Rhubarb, forsythia, Dandelions, Borneol and Calcined Gypsum composed three groups and were made as plasters.Through detecting these indicators such as general observation, White blood cell and platelet count, hyaluronic acid, Vitro experiments, filter out the best effect of promoting wound healing.Then continue to study the best prescription, Through detecting these indicators such as histopathological observation,Des hydroxy acid, NO, cytokines content and the expression level of TGF-β1mRNA, etc.to further explore the mechanism of chuangshangyuhe ointment promoting wound healing. The results show:1 Blood leukocyte and platelet content of rats were increased after the Fangfushengji ointment application to rat wound.and effect of antibacterial were markedly.That showed fangfushengji ointment could inhibit the growth of wound bacteria,clean-up of necrotic tissue,enhance partial immune function.2 Neutrophilic leukocyte and lymphocytes were activation by The fangfushengji ointment,made them chemotaxis to the wound, and immunologic active material were released,such as macrophage chemotactic factor , partial immune function were enhanced.3 The fangfushengji ointment could enhance hyaluronic acid and NO content in wound tissue or serum, and lower content of hydroxyproline in wound tissue.It showed that fangfushengji ointment not only promoted collagen synthesis and proliferation of granulation tissue, but also prevent scarring because of excessive synthesis of collagen.. 4 The Fangfushengji ointment could enhance IL-1β,IL-8,TGF-β1 protein content and expression of TGF-β1mRNA volume in Wound tissue,it showed that Fangfushengji ointment could regulate the content of cytokines, promoted inflammatory cell infiltration of local injury, directly or indirectly induced the formation of capillary endothelial cells, promoted granulation tissue formation, fibroblast-induced chemotaxis and activation, produced extracellular matrix protein.5 After observed the wound tissue microstructure and ultrastructure we found that fibroblasts increased, rich in the cytoplasm, capillary number increased and collagen fibers were arranged regularly in Fangfushengji group, they showed that the Fangfushengji ointment could promote organize angiogenesis in wound, improve local wound microcirculation stasis status, increasing local blood supply, accelerate wound metabolism, promote collagen synthesis, thereby promoting wound repair.
