Relationship Among Change Of Rap1GAP Gene And Protein In Cervical Cancer And HPV Infection

Objective: To reveal the molecular mechanism of cervical cancer by exploring the relationship among change of Rap1GAP gene and protein in cervical cancer and HPV infection.Methods: Collected 20 cases with normal cervix and 20 cervical cancer, HPV infection were detected by immuno-histochemistry for 40 samples; extracted DNA from the samples , Deletion of Rap1GAP exon 11 and Rap1GAP exon 19 were identified by polymerase chain reaction(PCR) amplification; extracted RNA by Trizol method, Expression of Rap1GAP mRNA was evaluated by reverse transcription polymerase chain reaction (RT-PCR);Expression of Rap1GAP protein was detected by immunohistoch emistry .Results:1. In 40 samples ,The HPV-positive was 19 cases(47%), The HPV- negativeWas 21 cases(53%),Infection rate of HPV was 20%(4/20)in normal cervix75%(15/20)in cervical cancer.2. Deletion rate of both Rap1GAP E11and Rap1GAP E19 was 0(0/20)innormal cervix, 10%(2/20)in cervical cancer.And there are no differences between them(P>0.05).3. Positive expression and relative content of Rap1GAPmRNA was detected less in cervical cancer(15%,0.721±0.387)than normal cervix (100%,1.428±0.470),there are significant differences between them(P﹤0.05).4. Positive expression rate of Rap1GAP protein was 15%(3/20),weak positive Positive rate 15%(3/20),negative rate70%(14/20)in cervical cancer.In normal cervix, positive expression rate55%(11/20), weak positive nagative rate45%(9/20),compared them P﹤0.05.5. Deletion of E11and E19 has no different between HPV positive samples and HPV nagatives in cervical cancer(P>0.05),Expression rate of Rap1GAP mRNA was 0(0/15) in cervical cancer with HPV positive and 60%60%(3/5)with HPV nagative, there are significant differences between them P﹤0.05. In HPV positive cervical cancer, Positive expression rate of Rap1GAP protein was 13%(2/15),weak positive Positive rate 7%(1/15),negative rate 80%(12/15),less than HPV- negative samples , there are significant differences between them(P﹤0.05).Conclusion:1.There was no deletion of both Rap1GAP E11and Rap1GAP E19 in cervical cancer.2.Expression of Rap1GAPmRNA and Rap1GAP protein were less in cervical cancer,It may relects the progression of disease.3. There was positive correlation between HPV infection and down- regulated expression of Rap1GAP , No correlation between HPV infection and deletion of both Rap1GAP E11and Rap1GAP E19.
