ObjectiveResearch correlation Rap1 GAP with invasive breast cancer, provide the basis of molecular biology in study of the pathogenesis of the latter for further, and to explore the relationship between Rap1 GAP expression and invasive breast cancer prognosis.MethodsA analysis by immunohistochemistry of the Second Affiliated Hospital of Fujian Medical University, the expression of 136 cases of invasive breast cancer and 15 cases of benign breast disease surgical specimens, 10 cases of normal breast tissue in paraffin sections Rap1 GAP, and on invasive breast Rap1 GAP expression with age, menstruation, receptors( including ER, PR, Her2), the tumor size, TNM stage, histological grade and aspects of axillary lymph node metastasis, and other single factor analysis, screening meaningful factors, then the binary multivariate logistic regression analysis. Disease free survival and overall survival as the evaluation of patients prognosis. Kaplan-Meier method to compare survival curves plotted, the prognosis for survival analysis using log-rank test, conducted survival differences, evaluation Rap1 GAP expression and invasive breast cancer prognosis.Results1.Rap1 GAP expression of invasive breast was 32.35%( 44/136), significantly lower than 60%(9/15) of the normal breast tissue and 90%( 9/10) of benign breast disease, the difference was statistically significant(χ2=16.299, P=0.000). SP staining showed under light microscope, Rap1 GAP expression in normal breast tissue and benign breast disease mainly localized in the cytoplasm, the degree of staining deep, strong positive or moderately positive; rather shallow extent of invasive breast cancer cells staining or no staining, weakly positive or negative.2.Univariate analysis showed that, Rap1 GAP expression have no relevant with age, menstruation, tumor size, receptor(ER, PR, Her2),difference was not statistically significant(P>0.05), have relate with TNM stage, histological grade and axillary lymph node metastasis, Rap1 GAP positive rate in TNM staging I-II period was higher than in phase III(P<0.05), Rap1 GAP expression of the organization grading I-II level was higher than level III(P<0.05), Rap1 GAP expression in that without axillary lymph node metastasis was higher than that with axillary lymph node metastasis(P<0.05). The differences were statistically significant.3.Multivariate Logistic regression analysis showed that TNM stage, histological grade and axillary lymph node metastasis were risk factors of Rap1 GAP expression(OR=2.689,4.017,2.567,P<0.05).4. Prognostic survival analysis showed, ① patient with Rap1 GAP expression of the three-year disease-free survival was 79.9%, five-year disease-free survival rate was 69.4%; three-year overall survival rate was 91.7%,five-year overall survival rate was 77%. ② patient with Rap1 GAP negative expression of three disease-free survival was 57.4%, five-year disease-free survival rate was 45.4%; three-year overall survival rate was 77%, five-year overall survival rate was 60.2%. By the log-rank test, there was a significant difference in survival of two groups of patients(P<0.05),Rap1GAP expression in patients of DFS and OS were significantly better than patients with negative expression Rap1 GAP.ConclusionRap1GAP expression in invasive breast reduction, suggesting that the mechanism of Rap1 GAP in tumor inhibition plays a role in cancer, TNM stage, histological grade and axillary lymph node metastasis have risk factors for the decrease in Rap1 GAP expression. Rap1 GAP expression than patients positive prognosis is relatively good, Rap1 GAP expression can be used to predict the risk of invasive breast cancer recurrence and prognosis...
