In this subject we research the influences of Pixu and Jianpi drugs on mucus secretion in rat model of COPD, and observe the influences of Pixu and Jianpi drugs to indicators of immune -inflammation, oxidation, nutrients, apoptosis, and explore mechanisms of"Pi as the source of sputum"from the points of immune-inflammatory, oxidation, nutrition, apoptosis, to reveal the essence of"Pi as the source of sputum",to provide new ideas and theory for understanding of the classic theory, guiding clinical practice, expanding the scope of application.In the experiment,48 rats were randomly divided into 4 groups: blank control group, COPD model group, COPD with Pixu model group caused by rhubarb, Jianpi treatment group with Liujunzi Tang.We use smoke and tracheal instillation of lipopolysaccharide to copy COPD model, use rhubarb gavage to copy pixu model, use Liujunzi Tang gavage as method of Jianpi, 36 days later,gather the specimen.1. General situation,such as rats cough, asthma, nose and mouth secretions, activity status, hair gloss, waste, appetite, body weight;2.Body weight changes during the experiment;3.The appearance of lung tissue; 4.Experiment of phenol red excretion to observe the secretion of bronchial sputum; 5.Collection of bronchoalveolar lavage irrigation lotion and cell sorting; 6.The spleen index; 7.HE staining to observe lung tissue pathological changes;8.Immunohistochemical measurement of MUC5AC expression in lungtissue;9.Radioimmunoassay measurement of IL-2, TNF-αin lung tissue homogenate; 10.Radioimmunoassay measurement of MDA in blood; 11. TUNEL measurement of apoptosis cells in lung tissue. Experimental datum are analyzed by SPSS11.5.We take phenol red excretion and MUC5AC expression in lung tissue as lung sputum indicators ,take TNF-a of lung tissue homogenate and cell sorting of bronchial alveolar lavage fluid as lung inflammation indicators,take spleen index and IL-2 of lung tissue homogenate as indicators of pulmonary immune status,take body weight changes as indicators of nutritional status.Compared with COPD model group, indicators of Pixu COPD model group have significant differences, indicators of Jianpi treatment group have varying degrees of differences.The results manifest that there are airway mucus hypersecretion phenomenon for COPD model.Pixu can aggravate mucus hypersecretion;Jianpi medicine Liujunzi Tang can significantly reduce the airway mucus hypersecretion.And immune-inflammation indicators,oxidation indicators,nutrition indicators,apoptosis indicators are significantly affected,suggesting that pixu and Jianpi medicine can affect airway mucus secretion through mechanisms of immune-inflammation, oxidative stress, nutrition and apoptosis. Neural mechanism, allergy mechanism, gastroesophageal reflux and a number of special food also participat in the mechanisms that "Pi pathological changes can affect Fei and sputum disease of Fei" .The above can be taken as the essence of "Pi as the source of sputum " in Fei diseases.
