ObjectiveBronchial asthma is a common disease in clinic.It is also a common disease in respiratory system.Recent years,economic in our country developed quickly,and the prevalence rate of bronchial asthma increased significantly.It is very hard to cure asthma and the disease often reoccurs.So it often influences the physical and mental health of patients and get enormous burden to society.It is an important topic for modern medicine on how to discuss the mechanism asthma and how to find useful method to prevent and cure asthma.In our study,asthmatic guinea pigs sensitized by ovalbumin were experimental model,and eosinophil, endothelin-iand transforming growth factorβ1 were observation indexes. We discussed about the anti-inflammatory mechanism of point application therapy on bronchial asthma at the cell and molecular level,which provided scientific theoretical basis for point application therapy in treating with bronchial asthma.MethodsForty guinea pigs were randomly divided into four groups,(normal control group,model group,dexamethasone group and acu-point application group),and each group has ten guinea pigs equally.The last three groups were got successful modeling.The method of making model was ultrasonic atomized inhalation of OVA.Acu-point application group:Selected herb: Ephedra,Seeds of Brassica alba,Asarum and Euphorbia kansui.Selected points:Dazhui(DU14),Feishu(BL13),Shenshu(BL23).Six hours for each time,once two days,seven times totally.The guinea pigs in dexamethasone group were injected dexamethasone(0.5mg/kg) in the abdomen,and the course of treatment was as the same as acu-point application group.The guinea pigs in model group recovered naturally.The guinea pigs in model group grew naturally with no any intervention.TGF-β1 in serum after experiment were determined by ELISA.The changes of the shape and structure of bronchia and lung tissues were observed by microscope.The expression level of ET-1 and the infiltration of EOS in bronchia and lung tissues were detected by immunohistochemical method(SABC staining).All the results were scanned and semi-quantitatively analyzed by computer image.Statistical analysis was carried out using SPSS 13.0 for windows.Results1.Influence of acu-point application on the EOS infiltration in bronchia and lung tissues of asthmatic guinea pigsThe number of Eos in lung organization goes up obviously in the model group and it has obvious difference from the number of the normalgroup (P< 0.01);The number of Eos in lung organization in the dexamethasone group and point application group are lower than the modelgroup(P<001); And the numbers have obvious difference from them(P<0.01).2.Influence of acu-point application on the ET-1 protein expression in bronchia and lung tissues of asthmatic guinea pigsThe ET-1 positive cell counting in bronchia and lung tissues of modelgroup was obviously higher than that in the normal control group (P<0.01).The ET-1 positive cell counting of dexamethasone group and point application group was significantly lower than that in the modelgroup (P<0.05).3.Influence of acu-point application on the TGF-β1 in the serum of asthmatic guinea pigsin the serum of the model group,the TGF-β1 evel is higher than that of the normal control group(P<0.05).In the serum of the acu-point application group and the dexamethasone group,the TGF-β1evel is lower than that of the model group(P<0.05).There was no difference between dexamethasone group and acu-point application group(P>0.05).Conclusions 1.There were obvious EOS infiltration in the bronchia and lung tissues of asthmatic guinea pigs sensitized by ovalbumin.EOS infiltration plays a important role in pathogenesis of allergic asthma.2.Acu-point application and dexamethasone injection therapies can reduce EOS infiltration and inhibit the ET-1 expression in the bronchia and lung tissues of asthmatic guinea pigs,and decrease serum content of TGF-β1.These result indicated that acu-point application and dexamethasone injection therapies anti airway inflammation and reduce airway hyper-reactivity by inhibiting local expression of ET-1 and adjusting the TGF-β1 content in peripheral blood.3.Acu-point application therapy can adjust the inflammatory reaction and control asthma and ameliorate the lesion of lung,so as to reach the treatment purpose.
