| Partâ… Expression and significance of LUNX and CK19 in non-small cell lung cancerBackground and aims:Since traditional RT-PCR technology is at best Semiquantitative,it has been difficult to differentiate between baseline level of gene expression in normal tissues and increased level of gene expression associated with cancer,rising the concerning for false-positive results.This study aimed to set up a quantitative method(i.e.real time fluorescent quantitative RT-PCR,Real-Time RT-PCR) and detection of NSCLC and lymph node tissues of cancer,NSCLC patients in order to accurately provide accurate staging and treatment of the theoretical basis. Methods:Using LUNX mRNA,CK19 mRNA as a marker,detected in 40 tumor tissues and 146 regional lymph nodes taken from 40 patients with NSCLC,16 lymph nodes from 11 patients with pulmonary benign diseases as controls by quantitative analysis with reverse transcriptase polymerase chain reaction(Real-Time RT-PCR). Meanwhile,all lymph nodes were also examined by conventional pathological method.Results:â‘ the mRNA level of LUNX in cancer Tissue was significant higher than that in control group,but has no association with Lymphatic Metastasis, Tumor Size and TNM grade.â‘¡the mRNA level of LUNX in the cancer tissues was significant higher than which in control group,and is associated with Lymphatic Metastasis,Tumor size and TNM grade.â‘¢the mRNA of CK19 in lymph nodes is almost as same as control,and not associated with tumor Size;Related to Lymphatic Metastasis and TNM grade.â‘£the mRNA of LUNX in lymph nodes is almost as same as control,which has no association with age,sex and histological type,related to Lymphatic Metastasis,Tumor Size and TNM grade.⑤mRNA of CK19 and LUNX is unrelated to age,sex and histological type.â‘¥there is Significant difference between using Real-Time RT-PCR methods and the routine pathological methods to detect lymph node metastasis in lung cancer.Conclusion:The results suggest that the Real-Time RT-PCR detection of LUNX message RNA and CK19 message RNA might be helpful to diagnose NSCLC micrometastasis in lymph nodes.LUNX is superior to CK19 both in sensitivity and specific.The establishment of this method may lead to positive rate of metastasis of lung cancer and treatment of patients with subclinical metastasis in regional lymph nodes.Partâ…¡Expressions of LYVE-1,CD31 in NSCLC and the correlation with neoplasm metastasisBackground and aims:The invasion and metastasis of neoplasm is constituted by some complicated and multi-step procedure,which related with the adhesion, angiogenesis,lymphatic metastasis and hematogenous metastasis of the tumor cells. NSCLC is to ensure that the principles of radical surgery and improve the survival rate of patients.Recent years,with the discovery of many lymphatic endothelial and vascular endothelial markers.The tumor angiogenesis and lymphangiogenesis research gradually increase.This paper detects lymphatic endothelial cell-specific markers LYVE-1 and vascular endothelial cell markers in CD31 expression in NSCLC,to explore its relationship with blood vessels and tumor lymphangiogenesis and metastasis.Methods:LYVE-1 and CD31 are detected by EnVisionTM immunohistochmistry method in tumor tissues and adjacent tissues from 40 patients; the expression of LYVE-1 and CD31 is used for the mark of the lymphatic endothelial cell and vascular endothelial cell,the Lymphatic Vascular Microvascular Density (MLVD) and microvascular density(MVD).Results:The MLVD of tumor tissues,normal lung tissues and adjacent tissues are significantly in NSCLC(P<0.01);the MLVD of the lymph node metastasis and without lymph node metastasis were significantly(P<0.01);MLVD are significantly withâ…¢+â…£stage thanâ… +â…¡stage in TNM stage(P<0.01);There are no significantly age,sex,tumor size,histological type and differentiation in NSCLC tumor tissues(P>0.05).The MVD of tumor tissues is significantly higher than adjacent tissues(P<0.01);The MVD of lymph node metastasis is higher than in those without lymph node metastasis,difference being not statistically significant(P>0.05);The MVD ofâ…¢+â…£stage is higher thanâ… +â…¡stage in TNM stage,difference being not statistically significant(P>0.05); analyzing the relativity,there was no correlation MLVD/MVD in tumor of NSCLC(P>0.05).Conclusion:NSCLC in the LYVE-1 and CD31 expression levels could be used as the disease progress and one of the indicators of tumor metastasis. |
