Objective:Lung cancer metastasis related protein 1(LCMR1) was a novel gene cloned by differential display reverse transcript PCR(DD-PCR) between human large-cell lung cancer cell lines 95C and 95D with different metastatic abilities.To explore the relationship between the expression of LCMR1 protein and the clinic features,overall survival(OS) and progression-free survival(PFS) of NSCLC patients respectively,we investigated the expression of LCMR1 protein in primary tumor tissues,tumor-adjacent tissues and metastatic lymph node of NSCLC patients by immunohistochemical staining.Methods:Immunohistochemical staining was used to investigate the expression of LCMR1 protein in 84 cases of paired primary tumor tissues and tumor-adjacent tissues,and 51 cases of metastatic lymph node of NSCLC patients by rabbit anti-human LCMR1 polyclonal antibody.All patients received neither chemotherapy nor radiotherapy before surgery.Immunohistochemistry results were determined by microscope analysis.We analyzed the relationship between the expression of LCMR1 protein and patients' gender,age,pathological types,smoking status,clinical stage,metastasis of lymph node,differentiation,OS and PFS respectively.All statistical analyses were carried out with the SPSS15.0 statistical software package.Results:(1) The LCMR1 protein was mainly located in the cellular cytoplasm;(2)The expression of LCMR1 protein was significantly higher in primary tumor tissues and metastatic lymph node of NSCLC patients than tumor-adjacent tissues;(3) The expression of LCMR1 protein in primary tumor tissues had a significant relationship with the clinical stage of NSCLC patients,and no relationship with gender,age,pathological types,smoking status,metastasis of lymph node,differentiation of patients.(4)The expression of LCMR1 protein in tumor-adjacent tissues had no relationship with the clinic features of NSCLC patients;(5) The expression of LCMR1 protein in metastatic lymph nodes had no relationship with the clinic features of NSCLC patients;(6) The expression of LCMR1 protein in primary tumor tissues was not significantly associated with OS and PFS of patients using single factor analysis and COX proportional hazard model analysis.Conclusions:The relationship between the expression of LCMR1 protein and clinic features of NSCLC patients was explored at the protein level.The results showed that the expression of LCMR1 protein was higher in primary tumor tissues and metastatic lymph nodes than tumor-adjacent tissues.In addition, the expression of LCMR1 protein in primary tumor tissues had close relationship with clinical stage.This suggested that LCMR1 could be a proto-oncogene that was playing a vital role in the carcinogenesis of NSCLC.
