Background:Early diagnosis and treatment play an important role in the prognosis of lung cancer patients in the process of diagnosis and treatment.The disintegrin-metalloproteinase family(ADAMs)is a transmembrane protein that contains a domain of disintegrin and metalloproteinase.It may have cell adhesion and protease activity.They play an important role in regulating cell phenotype by affecting cell adhesion,migration,protein hydrolysis and signal transduction.Functional ADAM participates in exogenous exfoliation of various growth factors,cytokines,receptors and adhesion molecules.Some ADAMs play a role in spermatogenesis and sperm function,possibly by affecting sperm maturation and adhesion and migration in uterus.Other non-catalytic ADAMs function in the central nervous system through the influence-oriented mechanism.Therefore,it is not surprising that ADAMs are the basis of many control processes in development and homeostasis,and when they are dysfunctional,they are also associated with pathological conditions(including cancer,cardiovascular disease,asthma,Alzheimer's disease).Similarly,studies have shown that ADAM plays animportant role in the invasion and metastasis of malignant tumors.Among them,ADAM12,also known as fusin-alpha,is also a member of the ADAMS family.It was initially discovered as one of three molecules in mouse myoblast fusion.There are two splicing variants: human ADAM12 includes transmembrane(ADAM12-l)and secretory(ADAM12-s).ADAM12 has many functions that contribute to protein hydrolysis,cell adhesion,cell fusion,signal of exogenous loss of ligand and cell apoptosis.Therefore,ADAM12 is involved in various pathological conditions.Overexpression of ADAM12 has been reported in breast,bladder,liver,lung,stomach and colon related malignant tumors,glioblastoma,giant cell tumors of bone and malignant tumors of blood system.However,there are few studies on non-small cell lung cancer(mainly lung adenocarcinoma and squamous cell lung cancer)and few clinicopathological analyses.Although all ADAMS contain metalloproteinase domains,only 13 of the 21 genes in humans encode functional proteases,suggesting that protein-protein interaction is a key aspect of their biological functions for at least eight other members.Functional ADAM participates in exogenous exfoliation of various growth factors,cytokines,receptors and adhesion molecules.Among them,ADAM12,also known as fusin-alpha,is also a member of the ADAMS family.It was initially found as one of three molecules in mouse myoblast fusion.There are two splicing variants: human ADAM12 includes transmembrane(ADAM12-l)and secretory(ADAM12-s).ADAM12 has many functions that contribute to protein hydrolysis,cell adhesion,cell fusion,signal of exogenous exogenous loss of ligand and cell apoptosis.Therefore,ADAM12 is involved in various pathological conditions.Overexpression of ADAM12 has been reported in breast,bladder,liver,lung,stomach and colon related malignant tumors,glioblastoma,giant cell tumors of bone and malignant tumors of blood system.However,there are few studies on non-small cell lung cancer(mainly lung adenocarcinoma and squamous cell lung cancer)and few clinicopathological analyses.Objective: To detect the level of ADAM12 in lung tissue of patientswith lung cancer,and to determine whether there is overexpression of ADAM12 in non-small cell lung cancer;compare with the corresponding clinical data,to explore the relationship between the expression of ADAM12 in lung tissue of lung cancer patients and the invasion and metastasis of lung cancer patients.Methods: This project recruits lung cancer patients who are scheduled to undergo surgery in Zhejiang People's Hospital from August2014 to November 2017.Postoperative pathology confirmed the diagnosis,which accorded with the diagnostic criteria of NSCLC.TNM stage was I-IV.Those who had not received preoperative radiotherapy or chemotherapy and had no other tumors except lung cancer.There were 30 cases of squamous cell carcinoma and 30 cases of adenocarcinoma.The expression of ADAM12 protein in lung cancer group and its adjacent tissues was detected by real-time quantitative PCR and immunohistochemistry.The relationship between ADAM12 protein expression and related imaging manifestations and clinical symptoms was analyzed.Statistical processing: SPSS13 software was used to analyze the results.Pearson F test or Fisher exact probability test,t test or t test,test and rank conversion non-parametric test were used according to the different objects.The test criteria were P < 0.05.Result: 1.ADAM12 is overexpressed in non-small cell lung cancer.2.The expression level of ADAM12 was not correlated with the histological type of NSCLC,but was significantly correlated with histological grade,clinical stage and lymph node metastasis.3.The expression level of ADAM12 increased with the increase of histological grade and clinical stage of non-small cell lung cancer patients.
