Objective:To observe and investigate the effect and the mechanism of Chinese herbal compound Bushenyifeijiedu recipe on rats with induced lung cacinoma as well as lung-kidney vacuity rats with induced lung caner,in order to provide scientific evidence for the clinical application of Bushenyifeijiedu(BSYFJD)recipe on the prevention of lung carcinoma especially lung-kidney vacuity type.Methods:72 SPF Wista rats,weighing 120~150g.Rats were divided randomly by 6 groups: disease model group,high dose of BSYFJD for disease group,low dose of BSYFJD for disease group,diease patten model group,high dose of BSYFJD for disease patten group, low dose of BSYFJD for disease patten group.Each group had 12 rats.Rats' model of induced lung carcinoma was set up by intra-left lobar-bronchial instillation with carcerogen. Mouse model of lung-kidney vacuity type was set up by hanging,smoking,swimming.The effect of BSYFJD recipe on normal condition of rats with induced lung carcinoma as well as lung-kidney vacuity rats with induced lung cancer was observed.Weight of lung and spleen was measured.Every stage of canceration of lung was observed by making pathological section Expression of oncogene c-mycmRNA and cancer suppressor gene p16mRNA was assayed by RT-PCR.Results:After inducing some rats appeared lung carcinoma while some appeared atypical hyperplasia of bronchus.The incidence of lung carcinoma(including precancerous lesion) in model group was higher than drug group.The spleen index of model group was lower than drug group while the lung index of model group was higher than drug group,especially for the lung-kidney vacuity type.BSYFJD recipe could significantly inhibit the incidence of inducing lung carcinoma and inducing lung-kidney vacuity type lung carcinoma on rats compared with disease group and disease patten group(P<0.05).It could release the pathological condition of lung,protect the immune organ and improve the rats' immunity function,particularly for the high-dose group(P<0.05).Compared to disease model group, high-dose BSYFJD recipe could decrease the expression of c-mycmRNA and inhibit the loss of the expression of p16mRNA(P<0.05) of cancerous tissue of rats with inducing lung cancer.Compared to disease patten model group,neither high nor low dose BSYFJD recipe could inhibit the over expression of c-mycmRNA(P>0.05) of cancerous tissue of lung-kidney vacuity rat with induced lung caner,but it could inhibit the loss of the expression ofp16mRNA(P<0.05).Conclusion:Lung-kidney vacuity syndrome may enhance the pathological degree of lung by decreasing the immunity function of rats.Bushenyifeijiedu recipe could significantly inhibit the incidence of lung carcinoma on rats.The effect of BSYFJD recipe on lung-kidney vacuity rats with induced lung cancer was also significant.The related mechanism may be that BSYFJD recipe could improve the immunity function of rats,release the pathological degree of lung and decrease the expression of c-myc as well as inhibit the loss of the expression of p16.
