Depression is the common type of mood disorders,it is the syndrome with features of significant and long time lasting decline in the mood.Its pathogenesis is not clear,and the treatment of depression is still built on the basis of the hypothesis,it is difficult to achieve a satisfactory effect.At present,the research emphasis is on the pathogenesis of depression,and developing better efficacy,and less side effects new drugs.1.Modeling of chronic unpredictable mild stress(CUMS) depressionModel rats will be used the method of raised alone,and accept the 21-day unpredictable stress to observe the changes on rat body weight,appetite and the impact of behavior.The experimental results show that:appetite of the model group rats was significantly lower than the control group(P<0.05),weight was significantly lower(P<0.01),and the level of vertical and horizontal movement significantly reduced(P<0.01).2.The research on neurotransmitters'change of CUMS depression model rat in different encephalic regionsUsing HPLC-electrochemical method decides neurotransmitter content of the depression model rat.The results showed that:the hypothalamus,striatum, hippocampus 5-hydroxytryptamine(5-HT),norepinephrine(NE) and dopamine (DA) of the depression model rat and its metabolites were significantly lower (P<0.01),and in some of them the relationship between each other is in disorder.3.The pathologic changes in brain tissue of CUMS depression model ratThe 5-HT expression changes in cortical and hippocampal of CUMS depression model rat.Under light microscopy,it shows that 5-HT immunohistochemistry-positive cells are in the cortex of theⅠ-Ⅴlayer, hippocampus are mainly in the CA1,CA3 areas.5-HT positive cells exist in the cytoplasm,nucleus negative.Compared with normal group,rat cortical, hippocampal CA1,CA3 District 5-HT immunoreactive neurons number of model group decrease and the average optical density decreased,there was a significant difference(P<0.01).4.The effect of Shu Yu San to CUMS depression model rat4.1 The pharmacodynamic study of medicine Shu Yu San to CUMS depression model ratAfter modeling,the rats will be accept the 21-day Shu Yu San treatment to observe the drugs effect on rat body weight,appetite and the impact of behavior. The experimental results show that:Shu Yu San can help the rats fight chronic stress-induced depression,control the body weight at slow growth,and increase the level of horizontal movement(P<0.01),vertical movement(P<0.01) and the forced swimming activities numbers(P<0.01).4.2 The Shu Yu San influence on the neurotransmitters of the depression model ratsUsing HPLC-electrochemical method test the neurotransmitter content of depression model rats after Shu Yu San treatment.The experimental results show that:Shu Yu San can increase 5-HT,its metabolites and antecedent content(P<0.01).Conclusion:The Shu Yu San treating depression is related with improving the 5-HT,its metabolites and antecedent content,adjust 5-HT metabolism.4.3 The influence to the pathologic changes of CUMS depression model rats by Shu Yu SanThe cortex,hippocampus CA1,CA3 area and 5-HT immunoreactive neurons of Amitriptyline group and Shu Yu San group rats increased by a marked number, the average optical density increased significantly.Compared with the model group,there were significant differences(P<0.05).In summary,biological basis of depression is multi-neural.The levels and functions of monoamine neurotransmitter are the dominant factor of change.Through the research on treatment mechanism of Shu Yu San on depression,it was found that the possible treatment mechanisms of Shu Yu San are:increased brain levels of 5-HT antecedent,increased levels of 5-HT and improve communication disorder between the neurons caused by stress. Depression is the unity of the nerve-endocrine-neurotrophic multi-factors abnormal entity.And their roles are interrelated and mutually reinforcing,and the treatment of depression should be based on the whole,the overall situation,in order to achieve satisfactory results.
