ObjectiveTo measure the change of serum adipose triglyceride lipase(ATGL) and visceral adipose tissue-derived serpin(Vaspin) levels,analyzing the relationship of serum ATGL and Vaspin levels to metabolic and clinical biochemical indicators in the subjects with different glucose tolerances and weights,in order to explore the relationship of ATGL and Vaspin,which was discovered as adipokine recently,as well as the role in occurrence of T2DM and obesity.MethodsFasting serum ATGL and Vaspin levels were measured by Enzyme-Linked Immuno Sorbent Assay(ELISA) in 21 subjects of normal glucose regulation with normal weight(NW-NGR),27 subjects of normal glucose regulation with over weight or obesity(OW/OB-NGR),36 subjects of type 2 diabetes with normal weight(NW-T2DM) and 30 subjects of type 2 diabetes with over weight or obesity (OW/OB-T2DM).Meanwhile fasting true insulin(FINS),fasting blood glucose(FBG),hemoglobinA1C(HBA1C),bloodlipid,blood pressure, height,weight,waistline and hip circumference were measured,and waist to hip ratio(WHR),body mass index(BMI) were also calculated.Using homeostasis model assessment(HOMA-IR) estimated insulin resistance: HOMA-IR=FBG*FINS/22.5.Results1.Serum ATGL levels in T2DM(298.27±57.57ng/ml) was similar with that in NGR(307.23±54.12ng/ml)(P=0.36);The concentrations of ATGL in subjects with over-weight and obesity were significant lower than those in the normal-weight subjects(242.28±55.72 VS 363.23±49.51ng/ml,p<0.01);the concentrations of ATGL were significant higher in female than those in male(312.63±56.53 VS 225.25±47.55ng/ml) (P<0.01);Pearson relation analysis showed that serum ATGL concentrations were negatively correlated with BF%,BMI,WHR,TG and HOMA-IR(r=-0.271,r=-0.238,r=-0.375,r=-0.313,r=-0.164, p<0.05 or p<0.01),positively correlated with HDL-C(r=0.497,P<0.01);In a multiple stepwise regression analysis TG,WHR and BF%were independent variables of ATGL.2.Serum Vaspin levels in T2DM were similar with those in NGR(1.15±0.71 VS 1.07±0.52ng/ml)(P=0.41);The concentrations of Vaspin in subjects with over-weight and obesity were significantly higher than those in the normal-weight subjects(1.08±0.39 VS 0.53±0.11ng/ml p<0.01);the concentrations of Vaspin were significantly higher in female than those in male(0.98±0.23 VS 0.65±0.19ng/ml P<0.05);Pearson relation analysis showed that serum Vaspin concentrations were positively and significantly correlated with BF%,BMI,WHR,FINS and HOMA-IR(r=0.238,r=0.365,r=0.214,r=0.183,r=0.147 P<0.05或P<0.01);In a multiple stepwise regression analysis WHR,gender and HOMA-IR were independent variables of Vaspin.Conclusion1.Serum ATGL levels significantly lowed in subjects with obesity, ATGL has remarkable correlations withWHR,BF%and TG,suggesting a potential link between ATGL and lipid metabolism and obesity,ATGL may become another functionary target in preventing obesity.2.serum ATGL levels were significantly higher in female than in male,it was positively correlated with HDL-C,This is possible one of new pathogenesis of coronary heart disease lower in female than in male.3.Serum Vaspin levels significantly increased in subjects with obesity, Vaspin has remarkable correlations with WHR,HOMA-IR and gender, these reveal that Vaspin may contribute to the pathogenesis of diseases correlating with insulin resistance such as obesity and T2DM.
