Purpose: The current study found that, vaspin play an important role in obesity.It is closely related to obesity, type 2 diabetes,insulin resistance and inflammation. It can increase insulin sensitivity and improve the insulin resistance of type 2 diabetes mellitus. Based on the previous work, we want to study the mechanism of KuSuanTongTiao Formula based on the theory of Chinese medicine Indepth. To observe the relationship between the expression of vaspin expression which intervention related factors and the inflammatory pathway of IKKβ/NF-κB,in order to Discussion chemerin by inhibiting theinflammatory response to reduce insulin resistance. The results will be used for the pathological mechanism of insulin resistance of type 2 diabetes to provide new ideas, and to provide new targets and drugs for the treatment and prevention of type 2 diabetes.Method:1 ZDF rats being feed with hypsi-lipoids feeder for 12 months, in order to copy the obese rat model of insulin resistance of type 2 diabetes mellitus. The successful DM rats are randomly divided into DM model group, the group of KuSuanTongTiao,and the group of BiGeLieTong,and normal control group. All of the groups were Given the gavage for 12 weeks. To observe the general condition of rats,testing the blood glucose, blood lipids, Free fatty acid, insulin and glycosylated hemoglobin.2 Detecting the levels of serum MCP-1、TNF-α、IL-6、CRP and vaspin of all of the rats By the method of ELISA.3 The western-blot method and real-time flourescent quantitative polymerase chain reaction(PCR) technology were used to detect the expression of Vaspin、MCP-1、NF-κB and IKKβ.Result:1 The KuSuanTongTiao Formula can improve the general condition of diabetic rats, and can significantly reduce the weight.2 The KuSuanTongTiao Formula can reduce the level of FBG,HbA1 C,TC,TG and FFA.3 The KuSuanTongTiao Formula can reduce the level of inflammatory factors of vaspin, TNF-α, IL-6 and CRP.4 The results showed The KuSuanTongTiao Formula can reduce the expression of the Gene and protein of Vaspin, MCP-1,NF-κB and IKKβ in rats’ Adipose tissue.Conclusion: The KuSuanTongTiao Formula can improve the general condition of diabetic rats, and can significantly reduce the weight.Reduce the level of the FBG, HbA1 C, TC, TG, FFA,vaspin,TNF-α,IL-6 and CRP.Reduce the expression of the Adipose tissue of the fat tissue of diabetic rats with gene and protein.Also, it can reduce the expression of Gene and protein of Vaspin, MCP-1, NF-κB, IKKβ in rats’ Adipose tissue. It can regulate lipid metabolism, reduce weight, reduce inflammation, improve the sensitivity of insulin, improve insulin resistance.The KuSuanTongTiao Formula through the mechanism of regulating Decoction in the treatment of obese type 2 diabetes mellitus may be that vaspin is a bridge to inflammation factor, the disorder of lipid metabolism, obesity, insulin resistance of type 2 diabetes link.
