Sub-health is a state between health and disease.It is the performance for a certain period of time to reduce the vitality,function and adaptability of the symptoms subside,but incompatible with the modern medical-related diseases or sub-clinical diagnostic criteria.Chronic fatigue are one of the most typical and important performance in sub-health status.In China,white-collar workers over the age of 40 Class in the case of chronic fatigue more prominent of which 75%belong to sub-health population,40%for the chief complaint of chronic fatigue.Therefore searching for some effective measures has become an important problem which is in dire need to be solved.However,modern medicine there is no better present for its prevention and control measures by the diversity of sub-health symptoms and the difficulty of diagnosis.In view of the nature of TCM,it has an obvious advantage of control sub-health chronic fatigue.Early in the "Huangdi's Classic of Internal Medicine" in the description,there will be a discussion about fatigue.At present, TCM treatment of fatigue have also been many studies,and these supply the theoretical and experimental basis for study of TCM on sub-health.As the effect of TCM in the treatment of fatigue,their application will be more extensive.Even though its control of TCM has a clear effect,but is facing a huge crowd of sub-health fatigue,because of sub-health awareness is not high,and the decoction of Chinese medicine to take the inconvenience in order to limit the advantages of TCM to combat sub-health.It is necessary to find a simple and effective methods of prevention and treatment of sub-health in TCM.Disease in TCM based on differentiation,combined with the clinical features of sub-health and geographical characteristics of the Guangdong region,we have developed the er-zhi-wan herbal tea.This experiment includes two parts.The first part is based on the Chinese Medical Clinical Guide for Sub-health,5800 medical undergraduate students from one Guangdong medical university were surveyed by the self-made sub-health questionnaire.The second part is based on the pharmacology and experimental efficacy study of er-zhi-wan herbal tea:①Total flavonoid content as an index, investigate the clarifying effect of ZTC 1+1Ⅱclarifier in decoction of er-zhi-wan herbal tea.②To study the alleviative effects of er-zhi-wan herbal tea on physical fatigue on mice,we observed loaded swimming time,serum urea nitrogen,glycogen in liver and lactic acid of blood.ChapterⅠBackgroundObjective:Understanding of sub-health status,fatigue and the fatigue sub-health research.Methods:Chinese Journal Full-text Database and Chinese Biomedical Literature for the media,search the full text of academic journals which title contained the "sub-health","fatigue" and "sub-health fatigue",their analysis of literature review.Results:①Sub-health domestic scholars have done a lot of research and study,the current definition of sub-health and to determine the basic reference standard in October 2006 in the Chinese Medical Association published the "Chinese Medical Clinical Guide for Sub-health"②The past 5 years reported in the literature of the prevalence rate of sub-health vary greatly,between 20%-90%.As the sub-health research is limited to cross-sectional studies and case-control study,it can only be sub-health prevalence rate,and the incidence can not be.Thus the incidence of sub-health is still unknown.③Sub-health diverse clinical manifestations,but all parts of the country studies have shown that the frequency of fatigue is the first clinical manifestation.④Fatigue mechanism mainly related to energy substances, metabolites and environmental changes in the body changes and so on,which to us to study the fatigue sub-health laid the foundation for intervention.⑤Chinese medicine as early as the "Huangdi's Classic of Internal Medicine" in there is a fatigue-related descriptions.The treatment of fatigue have been many studies of TCM,for the treatment of fatigue sub-health made a theoretical and experimental basis.⑥Sub-health fatigue concept,the mechanism and intervention studies,such as the lack of a complete system conclusion.Conclusion:Sub-health against humanity has become the number one health killer invisible,sub-health state of fatigue is the most common manifestations,so it is imperative to intervene effectively sub-health fatigue.ChapterⅡThe clinical epidemiology study of sub-health fatigueObjective:The medical undergraduate students endure large stress because they face study,clinical practice and many life choices.To describe the distribution and prevalence rate of sub-health fatigue among them and explore the correlated factors, the study analyzed the sub-health fatigue of 5800 medical undergraduate students from one medical high school of Guangzhou.Methods:5800 medical undergraduate students from one Guangdong medical university were surveyed by the self-made sub-health questionnaire.The sub-health fatigue diagnosis was accorded with partial content of Chinese Medical Clinical Guide for Sub-health.Correlation factors were analyzed statistically using logistic regression method.Results:①A total of 5800 questionnaires were sent,5729 were recovered with the recovery rate of 98.78%,including 5599 valid questionnaires,with a qualified rate of 97.73%.Among the 5599 questionnaires,there were 101 undergraduate students (1.8%) of disease state were removed.Finally,5498 questionnaires were involved in the result analysis.②The total prevalence rate of sub-health fatigue in medical undergraduate students was 68.94%.The prevalence rate in female was higher than that in male(P=0.046).③The manifestations of sub-health fatigue mainly involved in often yawning,lack of resources to cope with daily life,easy flu etal.④The correlated factors included lack of security,paying no attention to food hygiene,lack of leisure activities,dissatisfying to health services,without sleeping and exercising.Conclusion:The prevalence rate of sub-health fatigue amongmedical undergraduate students is very high and the correlated factors are complicated.It is necessary to carry out health education for the students and effective intervention,prevention and treatment in many ways.ChapterⅢResisting fatigue effects of er-zhi-wan herbal teaExperiment 1Objective:To investigate the clarifying effect of ZTC 1+1Ⅱclarifier in decoction of er-zhi-wan herbal tea.Methods:①Using the ultraviolet spectrophotometry,the major factors which influence the clarifying process,including the quantity of clarifier,the concentration of decoction and the reaction time have been investigated based on the total flavonoid.②Er-zhi-wan prescription herbal medicine,add ten times the volume of water, decocted 2 times 2 hours,the combined filtrate,concentrated to 1:20(original drug: extract).Water extraction and alcohol precipitation method using 95%ethanol to liquid ethanol concentration of 90%,standing 24 hours.ZTC1+1Ⅱclarifier:according to product requirements configuration A,B components,will be 6%liquid volume of component B solution add liquid slowly trickle in,mix,and then 8%of liquid volume of component A slow trickle of add liquid,mix,and placing 60℃in a water bath holding 1 h,ambient temperature and then holding 30min,after 2000 r / min centrifugation 10min,the supernatant taken.Check the above-mentioned two methods of solution preparation and determination of stability of total flavonoid.Results:The optimized clarifying conditions are as follows:the amount of clarifier used is 8%for part A,6%for part B;the concentration of decoction is 1:20; temperature of 60℃reaction for 60 minutes.Conclusion:ZTC 1+1Ⅱclarifier can effectively remove the impurities of the decoction of TCM,and improve the content of total flavonoid(P=0.000);At the same time guarantee the quality of the stability of the solution(P=0.000).ZTC 1+1Ⅱclarifier agent applied to er-zhi-wan herbal tea,we can ensure that their active ingredients,but also to clarify the effect of reach.Experiment 2Objective:To study the alleviative effects of er-zhi-wan herbal tea on physical fatigue on mice.Methods:160 male Kun-ming mice were randomly divided according to body weight:normal control group,er-zhi-wan herbal tea group in low dosage,in middle dosage,and in high dosage.After the mice were orally administered er-zhi-wan herbal tea in low dosage,middle dosage and high dosage respectively for 30 days, loaded swimming time,serum urea nitrogen,glycogen in liver and lactic acid of blood were determined.Results:Er-zhi-wan herbal tea groups in middle and high dosage significantly prolonged the loaded swimming mean time of mice,while er-zhi-wan herbal tea group in high dosage obviously decreased serum urea nitrogen level after swimming.At the same time,it showed an evident incremental effect after swimming on glycogen in liver.The area average under the lactic acid of blood have the significance when compared the er-zhi-wan herbal tea group in high dosage and the normal control group.It is explained that high dose had reduces the movement mouse lactic acid of blood.Conclusion:These findings suggest that there were resisting fatigue effects of er-zhi-wan herbal tea on male Kunming mice.It provides a theoretical basis and experimental support for its development and utilization.But its mechanism of resisting fatigue remains to be further studied.
