Abstract/Summary:
Schizophyllum Commune is a rare large scale fungus which is traced back to civilization of Russia.It is widely used to prevent and cure various kinds of disease,especially cancer.Recent years,research on Pharmacologic activity and chemical composition of S. commune has been paid more attention to by researchers both at home and abroad.Artificial taming on large scale has not come true yet.The origin mainly depends on collection.This makes the development and exploitation of S.commune suffer confinement.While the fluid fermentation cultivation can overcome this deficiency.To gain mycelium by liquid submerged fermentation in this text,and research on Pharmacologic activity of mycelium and broth ofΙobliquus is carried on in this experiment.Bacteriostatic action,extraction technology of crude polysaccharide from mycelia,antitumor activity and mechanism of crude exopolysaccharide, antitumor activity of crude polysaccharide,anti-inflammatory action is determined,and extraction method of polysaccharide is determined as well.1.The direct results of this paper indicate that the optimization cultivation prescription of fluid fermentation of S.commune is:dextroglucose20g/L,maizena50g/L,KH2PO4 3.0 g/L,MgSO4·7H2O 1.5 g/L。The cultivation temperature is 28℃.The quantity of shake flask is 30%,the rate of rotation is 120r/min.The inoculation quantity of fluid strain is 12%.Mycelium dry weight is the highest on the 11th day of rotation.This result is similar to the previous two.This lay a foundation of further research on S.commune.2.Four bacteria(Salmonella enteritidis,Staphylococcus aureus,Salmonella typhimurium, Escherichia coli,) are used in this experiment to test the bacteriostasis action of mycelium and broth of S.commune by slip diffusion method and stipe method.The results show that mycelium and broth of S.commune can inhibit the growth of the four bacteria,This indicate that mycelium and broth of S.commune contain bacteriostasis active components,the bacteriostasis active components of mycelium is mainly in the extraction of n-butanol.3.Study on the optimized technology of polysaccharides extraction,using Microwave-assised extraction method,supersonic extraction method and hot water extraction method by orthogonality experiment respectively.The optimized condition of hot water extraction method is determined by analysis of range:Time is 4h.Times of extraction is 5.Ratio of fluid and powder is 50.Temperature of extraction is 90℃Extraction ratio of polysaccharide is 13.45%.Optimization technology of microwave extraction method:Time is 20min.Times of extraction is 4.Ratio of fluid and powder is 60.Extraction ratio of polysaccharide is 10.8%.Optimization technology of hypersound extraction method:Time is 40min.Times of extraction is 4.Ratio of fluid and powder is 60. Temperature of extraction is 50℃.Frequency of extraction is 70.Extraction ratio of polysaccharide is 19.53%.The proteoglycan in fermented liquid and mycelia were 8.76g/1000mL and 19.85g/100g (dride mycelia) separately which were extracted with supersonic and hot water,and precipitated with 95%alcohol.The content of polysaccharide is 79.42%and 66.63%in proteoglycan.4.Primary research on the tumor inhibitory action of crude exopolysaccharide of S.commune which is from solid tumor and ascitic tumor mice.Inhibition rate of sarcoma S180 and hepatoma H22 is primarily researched by giving tumor-bearing mice isotonic Na chloride(blank group), cyclophosphamide(positive control group),crude exopolysaccharide from S.commune of high dose(500mg/kg),medium dose(250mg/kg) and low dose(125mg/kg),respectively.High tunor inhibition activity and hypotoxicity characteristics of crude exopolysaccharide from S.commune is integratedly surveyed with the exponent of the thoracic gland and the exponent of the spleen as an index.The content of L-18 and TNF-αin the blood-serum of tumor-bearing mice is measured.Antitumor mechanism of action of crude exopolysaccharide fromΙ.obliquus is demonstrated briefly.The tumor inhibition rate of sarcoma S180 of medium dose group of crude exopolysaccharide from S.commune is 49.03%,low dose group is 36.41%.The tumor inhibition rate of hepatoma H22 of medium dose group of exopolysaccharide from S.commune is 51.54%, low dose group is 42.29%.high dose group can also reach 35.62%.The results exhibit that exopolysaccharide from S.commune can inhibit the growth of sarcoma S180 and hepatoma H22.And the exponent of the thoracic gland and the exponent of the spleen of medication administration team is obvious higher than bland group.This indicates that crude polysaccharide and crude exopolysaccharide from S.commune can enhance the organism immunity to some extent.The content of L-18 and TNF-a in the blood-serum of the two kinds of tumor-bearing mice is increased.This indicates that the antitumor role of crude exopolysaccharide from S. commune is played by enhanced the organism immunity when counteracting tumor.The research results of life elongation rate indicate that crude exopolysaccharide from S.commune can prolong the life time of tumor-bearing mice obviously.It can prolong life time of sarcoma S180 mice 1.88day on average.It can prolong life time of hepatoma H22 mice 3.78day on average.5.Antineoplasmic activity of crude polysaccharide from S.commune is tested by the same method as crude exopolysaccharide.The results show that inhibit effects of crude polysaccharide from S.commune of the three dose on sarcoma S180 and hepatoma H22 are obvious.The tumor inhibition rate of sarcoma S180 of high dose group of crude polysaccharide fromΙ.obliquus is 48.28%,medium dose group is 39.85%,low dose group can also reach 35.25%.The tumor inhibition rate of hepatoma H22 of high dose group is of crude polysaccharide from S.commune 40.71%,medium dose group is 37.50%,low dose group is 33.21%.The results exhibit that crude polysaccharide from S.commune can inhibit the growth of sarcoma S180 and hepatoma H22.And the exponent of the thoracic gland and the exponent of the spleen of medication administration team is obvious higher than bland group.This indicates that crude polysaccharide from S.commune can enhance the organism immunity when counteracting tumor.6.Inhibition effect on engorgement of mice induced by xylene is primarily researed by giving mice isotonic Na chloride(blank group),indomethacin(positive control group:10mg/kg), crude exopolysaccharide(polysaccharide)from S.commune of high dose(500mg/kg),medium dose(250mg/kg) and low dose(125mg/kg),respectively.The results show that crude exopolysaccharide and crude polysaccharide from S.commune possess obvious inhibition effect on engorgement of mice induced by xylene.Crude exopolysaccharide and crude polysaccharide from S.commune can enhance the immune competence of mice.The inhibition ratios of engorgement of mice of high dose,medium and low dose of crude polysaccharide from S. commune are 34.43%,22.68%,16.44%respectively.The inhibition ratios of engorgement of mice of high dose,medium and low dose of crude exopolysaccharide from S.commune are 20.01%,31.09%,12.90%respectively.
