Depression is the main type of mood disorders,which is the syndrome for the main features,of a significant and long-lasting depressed mood.It is take place in female frequently.It is onset at young-prime age.Following the development of the society,acceleration of the rhythm in working and living, the incidence rate of depressive disorder shows rising tendency year by year. Depressive disorder has become a emotional health problem in global.It has characteristics which is high attack rate,high recurrence rate,high burden rate and committed suicide rate.It has been common disease that is harm to people's physical and mental health,form huge burden and expenses to person,family and society.It is obscure about pathogenesis and etiology.We study the pathogenesis of depression from the angle of traditional Chinese medicine science.We think that the pathogenesis of depression is liver-Qi stagnation,and throughout the disease has always been.The occurrence of depression correlates with many elements,but Chinese herbal medicine emphasize unification and balance of whole,and treat complex syndrome, which shows unique superiority.In view of the superiority and the pathogenesis of depression,we used ancient prescription-Chaiqin Wendan Decoction(CWD) to intervene and further explored its mechanism. Objective To study effect of CWD on the behavior,Estradiol(E2), Neuropeptide Y(NPY) of depressed female mice induced by chronic unpredictable mild stress(CUMS).Method(1)Animal model was built with female mice by exposing the animals to a paradigm of chronic unpredictable mild stresses including 10 different stressors for a consecutive course of 3 weeks.(2) Open-Field test was performed to evaluate the capacity of CWD to improve depression-related symptoms,such as cognitive ability,spontaneous exercise to reduce and excitatory under CUMS model.(3) Detection by radioimmunoassay was performed to test levels of E2 in mice's serum and NPY in mice's plasma.Result(1) After 3 weeks of chronic stress on the mice,levels of crossing and rearing were reduced significantly.It is considered to be a more satisfactory model which can reflect the depressive illness.(2) After 3 weeks of CWD especially high doses of CWD administration(20g/kg),levels of crossing and rearing were significantly increased(P<0.01).And there were no significant difference between CWD and fluoxetine control group(P>0.05) in improving the depressed behavior.(3)The relevant indicators of depression such as E2 and NPY of CWD group were significantly enhanced(P<0.01). And the effect of high doses of CWD group was more significant than the low group(P<0.01).Compared CWD group to fluoxetine control group,E2 has a significant pick-up(P<0.01),while NPY was no significant difference(P> 0.05).Conclusion The statistical information of the experimental results show that CWD have the following functions:1.It can improve the depressed behavioral function of the depressed mice.2.It can increase E2 levels in the depressed mice's serum.3.It can increase NPY levels in the depressed mice's plasma.This research concludes that CWD is one kind of effective medicine which can prevent depression.It provides some experimental evidence for the treatment of depression...
