Objective: viral myocarditis is one disease caused by viral invaded heart, its main manifestation is cardiac muscle's inflammation. Its pathogenesis is not yet clear. At present many people think that virus'direct invasion is the main reason in the early. But later Immune pathological damage caused by virus or damaged heart is recognized by most people. In clinic, due to the lack of effective anti-viral drugs, on the basis of the conventional symptomatic treatment and support, non-hormonal anti-inflammatory drugs, Angiotensin converting enzyme inhibitors, Oxygen free radical scavenger and Immunosuppressant can not reach a satisfactory effect. As a traditional treatment, the advantages of traditional Chinese medicine in treating viral myocarditis has proved in clinic, and opens a new way for the treatment of VMC.Viral myocarditis is one of more common cardiovascular diseases in children. Intestinal virus, especially Coxsackie B3 virus (Coxsackie virus B3, CVB3) is the most common cause of VMC. It took my tutor's many years'clinical researches and combined the physiological and pathological features of children to set a new prescription---yinqiaoyimi decoction, and reached remarkable curative effect. This experiment use CVB3 duplicate mice's viral myocarditis model. By observing the influence of serum enzyme, SOD, MDA, TNF-α, organizational structure as well as cardiac cells caspase-3, explore the mechanism of yinqiaoyimi decoction in the treatment of viral myocarditis, and provide experimental basis for the clinical application.Method: selection of 60 male Balb/c mouse were randomly divided into five groups,each with 12, were gaps in the normal control group (which was abbreviated to"N group"), the model group(which was abbreviated to"M group").the ribavirin group (which was abbreviated to"R group"),the huangqi group (which was abbreviated to"H group"),the yinqiaoyimi decoction group, (which was abbreviated to"Y group").In addition to the"N group", the other four groups were inoculation liquid Coxsackie virus B3 Nancy 0.1ml,whith half of the tissue culture infection dose(which was abbreviated to TCID50)was 10-6.5/ml. At the same time the N group was treated with saline 0.1ml. After making model on mice one hour, Y group were dosed with 20g/kg, R group were dosed with 0.1g/kg, H group were injected with 0.2g/kg, N group and M group were dosed with saline 0.2ml/10g at the same time. Once a day, the total treatment was 10 days. After twenty-four hours of the last dose, all mice were decapitated and the blood were collected and centrifuged for reserve. Aseptic take heart, a part of them were fixed by 10% formalin, and then embedded by paraffin, then dying by haematoxylin and eosin, at last observing organizational structure of myocardial under the microscope. The other parts of the heart were fixed by 70% alcohol, after grated to detect Caspace-3 by using flow cytometry.Results:1 The general behavior of all the mice During the process of making model, the mice of N group was normal water and food consumption, the fur was luster, the activity was nimble, and having sensitive reflection; the rest of the mice have different degree of appetite lessens, loose fur, activity decreased and poor vigor. In which, M group was the heaviest, Y group was the lightest.2 The effects of yinqiaoyimi decoction on serologic index2.1 The effects of yinqiaoyimi decoction on myocardial enzyme2.1.1 LDH: Compared with N group, M group is significantly increased in LDH (P<0.01). Every treatment groups were decreased compared with M group (P<0.01). Among of all treatment groups, Y group is decreased compared with H group and R group (P<0.01), H group is also decreased compared with R group (P<0.01).2.1.2 AST: Compared with N group, M group is significantly increased in AST (P<0.01). Every treatment groups were decreased compared with M group (P<0.01), Among of all treatment groups, Y group is decreased compared with R group (P<0.01), H group is decreased compared with R group (P<0.01), Y and H group had no statistical significance (P>0.05).2.1.3 CK: Compared with N group, M group is significantly increased in CK (P<0.01). Every treatment groups were decreased compared with M group (P<0.01). Among of all treatment groups, Y group is decreased compared with R group and H group(P<0.01),H group and R group had no statistical significance(P>0.05).2.2 The effects of yinqiaoyimi decoction on SOD and MDA2.2.1 SOD: Compared with N group, M group is significantly decreased in SOD (P<0.01). Every treatment groups were increased compared with M group (P<0.01). Among of all treatment groups, Y group is increased compared with R group and H group (P<0.01),H group had no statistical significance compared with R group(P>0.05).2.2.2 MDA: Compared with N group, M group is significantly decreased in MDA (P<0.01). Every treatment groups were decreased compared with M group (P<0.01). Among of all treatment groups, Y group is decreased compared with H group and R group(P<0.05 or P<0.01),H group is decreased compared with R group(P<0.01).2.3 The effects of yinqiaoyimi decoction on TNF-αCompared with N group, M group is significantly increased in TNF-α(P<0.01). Every treatment groups were decreased compared with M group (P<0.01). Among of all treatment groups, Y group is decreased compared with R group and H group(P<0.01),R group is decreased compared with H group(P<0.01).3 The effects of yinqiaoyimi decoction on Caspase-3The contents of caspase-3 in the myocardium was expressed by fluorescence index (FI).The results show that compared with N group, M group is significantly increased in FI (P<0.01), Every treatment groups were decreased compared with M group (P<0.01).Among of all treatment groups, Y group is decreased compared with R group and H group(P<0.01or P<0.05),H group and R group had no statistical significance (P>0.05).4 The effects of yinqiaoyimi decoction on cardiac tissue structure of miceOf normal group, the myocardial fibers were arranged regularly, interstitial substance and vascular no changed, and no inflammatory cell and myocardial cell degeneration and necrosis. Of model group, the myocardial arranged disorder, lots of inflammatory cells, myocardial cell degeneration necrosis, and interstitial hyperemia and edema. Of all treatment groups, the myocardial fibers arranged orderly compared with model group, inflammatory cell infiltration, myocardial cell degeneration and necrosis significantly reduced, Y group is the lightest one.Conclusion:Yinqiaoyimi decoction can effectively reduce the serum enzymes of myocarditis mice, improve serum SOD content and reduce the content of MDA, inhibit the expression of TNF-α, reduce the content of Caspase-3 in myocardial and improve cardiac injury in mice.
