Effect Of HuaZhuoJieDu Decoction On The Level Of MTL,SS,ET In Rats Of Chronic Atrophic Gastritis

Objective:By observing the impact of HuaZhuoJieDu Decoction on chronic atrophic gastritis model rat's general situation,gastric mucosal morphology,MTL,SS,ET,explore the mechanism of HuaZhuoJieDu Decoction in treatment of chronic atrophic gastritis and lay the foundation for further study of HuaZhuoJieDu Decoction.Method: 60 healthy male Wistar rats randomly divided into five group:normal group,model group,WeiMeiSu group , Hua Zhuo Jie Du Decoction low dose group,HuaZhuoJieDu Decoction high dose group.Each group have 12 rats.Except the normal group,the remaining rats were modeled by integrated model law,total 90 days.Begin to treat after model been successfully build: high dose group use high dose HuaZhuoJieDu Decoction (7.14g crude drug/ml) irritate rats stomach once a day according to 71.4g/kg.d;low dose group use low dose HuaZhuoJieDu Decoction (2.38g crude drug/ml)irritate rats stomach once a day according to 23.8g/kg.d;WeiMeiSu Group use WeiMeiSu liquid(0.1g/ml) irritate rats stomach once a day according to 1.0g/kg.d.The equal saline was given to normal group and model group.All of the five group last 60 days.After treatment,all rats were killed to take blood sample and stomach.Then detect MTL,SS and ET in blood plasm through RIA.Observe pathology of gastric mucosa tissue by eye and light microscope. Measurement data are expressed with±s.Use SPSS16.0 statistical package for statistical analysis.P<0.05 mean the difference was significant.Result: (1)General condition of the rats:All the group of treatment can improve rat's fur color,the state of mind,the appetite,and the activity.But the high dose group has obvious influence.(2)Observe the gastric mucosa by eyes:Gastric mucosa surface of normal group are smooth, regular ,pink.The model group has the obvious pathological change.The mucous are pale and point shape or schistose bleeds are visible.Plica are low,rare and even disappear.WeiMeiSu group,low dose group and high dose group can improve gastric mucosa pathological change.The majority of high dose group are near normal.(3)Gastric mucosa under light microscope observation:Gastric mucosa thickness of normal group is normal.The epithelial cell is complete.The gland are arranged neatly.The blood capillary does not have the expansion.There have no inflammation react.The gastric mucosa of model group becomes thin.Epithelial cell have degenerated,necrosised and came off.Mucous glands have different degrees of shrinkage or disappearance.The mucous membrane skin layer is increased thick.Gastric mucosa of high dose group are moderate thickness.Epithelial cell have no significant degeneration and necrosis.The gland arranged close and regular.A small number of inflammatory cell infiltrated interstitum.The improvement of WeiMeiSu group and low dose group are not as good as high dose group.(4) The change of plasma motilin(MTL):Compared with the other groups,the normal group(80.49±8.56 pg/ml)have the low level of plasma MTL.The difference is significant(P<0.05).All of the treatment group can reduce the level of p l a s m a m o t i l i n . A m o n g W e i M e i S u g r o u p (97.88±4.87pg/ml),low dose group (96.21±3.56pg/ml),high dose group(90.88±3.87 pg/ml),the high dose group are the lowest(P<0.05).There is no significant difference between WeiMeiSu group and low dose group.(5) The change of plasma somafostatin(SS):Compared with the normal group ( 6 4 . 7 5±7 . 1 6 p g / m l ) , t h e p l a s m a S S o f model group (21.43±6.41pg/ml) ,WeiMeiSugroup(23.84±4.37),low dose group(22.41±5.46 pg/ml) and high dose group(40.88±5.07 pg/ml)are low.The difference is significant(P<0.05).The model group,WeiMeiSu group and low dose group have no significant difference(P>0.05).The difference between model group and high dose group is significant(P<0.05).The difference between low dose group and WeiMeiSu group is not significant(P>0.05).The difference between high dose group and WeiMeiSu group is significant(P<0.05).The difference between high dose group and low dose group is significant(P<0.05).(6) The change of plasma endothelin(ET):Compared with the normal group(70.27±7.61pg/ml),the plasma ET of model group(100.41±6.26pg/ml),WeiMeiSu group(88.35±3.48pg/ml), low dose group(80.89±4.21 pg/ml) and high dose group(75.95±3.31 pg/ml)are high.The difference is significant( P<0.05 ) .The difference between model group and WeiMeiSu group is not significant(P>0.05).The difference between model group and low dose group is significant(P<0.05).The difference between model group and high dose group is significant(P<0.05).The difference between high dose group and low dose group is significant(P<0.05).Conclusion:(1)HuaZhuoJieDu Decoction can improve general state of rat model of CAG.(2) HuaZhuoJieDu Decoction can improve impaired gastric mucosa of rat model of CAG. (3) HuaZhuoJieDu Decoction can adjust disordered level of MTL,SS,ET,increase gastric mucosal blood flow,improve microcirculation and enhance repair of mucosal barrier function.So that it can achive the aim of treating chronic atrophic gastritis.
