Objective:Establishing the cirrhosis rats model using the carbon tetrachloride synthesis method,the general condition,ETX,D-LA,DAO as main observation index,as well as observing the pathological changes of small intestine and liver in different groups,to explore the mechanism of Huazhuojiedu Decoction on improving the function of intestinal barrier in cirrhosis rat model,and to provide the experimental basis for the treatment of Huazhuojiedu Qingchang method on the treatment of cirrhosis.Method:48 clean wistar rats were selected,of which 8 were randomly selected as the blank control group?group N?,and the other 40 in the model group.Establishing the cirrhosis rats model using the carbon tetrachloride synthesis method?CCl4+edible alcohol+bean oil?.The successful model–established 26 rats were randomly divided into model group?group M?,Huazhuojiedu Qingchang Decoction group?group Z?,6 in each group;bifidobacterium triple viable enteric capsules?group X?,huazhuojiedu Qingchang decoction combined with bifidobacterium triple viable enteric capsules group?group C?,7 in each group.The rats in group N and M were given saline enema,and the rest groups were given corresponding treatment for 30 days.At the end of the experiment,the liver was completely stripped and the appearance was observed.The femoral artery was taken,the ETX level was detected by limulus reagent end point coloration,the reformed enzymatic method for the D-LA level,chemical rate method for the DAO activity.Meanwhile,the pathological changes of small intestine and liver by HE staining were observed.Results:1.The general situation of rats:the mortality of rats in this study was30%.In group N,rats were in good condition while the rats in model group represented the feature of rare fur,poor reactiveness and reduced amount of activity.With treatment,the general condition of the model rats got different degrees of improvement.2.ETX content:Compared with group N,the ETX content in group M was increased,the difference was statistically significant?P<0.01?;and the content of ETX in group C was lower than that in group X,the difference was statistically significant?P<0.05?;while group Z and group X had no statistical significant in ETX levels?P>0.05?.3.Levels of D-LA:the D-LA level of group M was higher compared with group N,while the D-LA level of Group Z and Group C was lower compared with group X.,the difference was statistical significant?P<0.05?,meanwhile,there was no statistical significant difference between Z group and C group?P>0.05?.4.DAO activity:the activity of DAO in group M was higher compared with group N,the activity of DAO in group Z and C was lower than group X,the differences were statistical significant?P<0.01?.However,in comparsion with group Z and C,there is no statistical significant difference?P>0.05?.5.Comparison of the appearance of liver:Group N:normal form of liver,smooth surface,bright color,complete capsule,soft texture;group M:the size of liver significantly reduced,hard,blunt edge and incomplete;group Z:complete capsule,red color,the surface was slightly granulate and is not hard in quality;group X:dark red,incomplete capsule,coarse grainy surface;group C:liver is smaller,light red in color,incomplete capsule,surface with fine granular,integrate edges.6.Histopathology of small intestine:group N:the intestinal villus were normal,arranged neatly,the epithelial cells were intact,no abscission and necrosis,and no infiltration of inflammatory cells ware found.Group M:the intestinal villus were arranged in disorder,most of the intestinal villus were broken and short,some of the villus were seen,the capillaries of the lamina propria were dilated and congested,and the focal infiltration of inflammatory cells was seen.The liver micromorphology of each treatment group was improvedin some level compared with group M.7.Liver histopathology:the size and shape of hepatocyte were normal in group N,and the structure of hepatic lobule was clear.In group M,the hepatocyte was damaged seriously,obviously fatty.Large numbers of fibrous tissues are lobulated hepatic lobules,forming typical lobular structures,inflammatory cells are diffuse infiltration.Compared with group M,the liver micromorphology of each treatment group were all have different degrees of improvement.Conclusions:1.Huazhuojiedu Qingchang decoction can obviously improve the function of intestinal barrier in cirrhosis rat model and improve intestinal mucosal injury.2.Huazhuojiedu Qingchang decoction can significantly improve the liver appearance of cirrhosis rat model,and reduce the degree of inflammation and hepatic fibrosis.3.The mechanism of huazhuojiedu Qingchang decoction on improving the function of intestinal barrier and liver function of cirrhosis rat model may be related to its function on decreasing the amount of plasma ETX,D-LA and DAO levels.
